Summary: The study explored the pathological role of a nonhistone methyltransferase in gastric cancer (GC), identified EEF1AKMT3 as a novel tumor suppressive methyltransferase, and revealed its mechanism of action through regulating the methylation of MAP2K7 and the degradation of TP53. The findings suggest potential targeted therapy in GC by manipulating the dysregulated EEF1AKMT3/MAP2K7/TP53 signaling axis.

Summary: The UTMD-mediated delivery of shRNA-Livin decreased survival and increased apoptosis of OVCA-433 cells, while also reducing the expression levels of caspase-3 and caspase-8. This study demonstrates that ultrasound mediation of targeted microbubbles can enhance the transfection efficiency of shRNA-Livin in ovarian cancer cells.

Summary: In this study, a near-infrared light-mediated mitochondrial RNA modification approach was developed for long-term imaging and effective tumor suppression. A tumor-targetable NIR fluorescent probe f-CRI was synthesized and it could covalently bind to mtRNA upon 808 nm irradiation, leading to prolonged retention in tumors. The covalent modification of mtRNA by f-CRI disrupted mitochondrial function and significantly suppressed tumor growth. This approach provides a potential strategy for cancer RNA interference therapeutics.

Summary: This study identified a novel lncRNA, LINC01348, significantly downregulated in HCC, which inhibited cell growth and metastasis by interacting with splicing factor 3b subunit 3 (SF3B3) to modulate EZH2 pre-mRNA AS. This novel regulatory association involving LINC01348/SF3B3/EZH2/JNK/c-Jun/Snail is a crucial determinant of metastasis in HCC cells, suggesting the potential utility of targeting LINC01348 as a therapeutic strategy for HCC.

Summary: The combination therapy of PDT and RNAi using ALHDL irradiation strategy effectively inhibits tumor growth and metastasis by improving VEGF silencing efficacy and angiogenesis inhibition. The use of ALS carrier system enhances tumor penetration and cellular uptake, leading to significant antitumor effects both in vitro and in vivo, while slowing down the rapid deterioration of the anoxic tumor environment.

Summary: The study showed that silencing of FANCF inhibits the function of the FA/BRCA pathway, resulting in inhibited cell proliferation, increased cell apoptosis, and DNA damage in OVCAR3 cells. Furthermore, silencing of FANCF enhances the antiproliferative effect of ADM in OVCAR3 cells, increases ADM intracellular accumulation, and sensitizes the cells to ADM.

Summary: The study identified a lncRNA called LINC00926 that negatively regulates PGK1 expression and predicts good clinical outcomes in breast cancer. Hypoxia inhibits LINC00926 expression and activates PGK1 expression through FOXO3A. This discovery established FOXO3A/LINC00926/PGK1 as a critical axis in regulating breast cancer growth and progression.

Summary: This study reveals an adaptive immunosuppression mechanism that leads to resistance to PARP inhibition in BRCA1-mutant ovarian tumors. This resistance is mediated by an increased population of protumor tumor-associated macrophages (TAMs) and activation of the STAT3 signaling pathway. The use of STING agonists can reshape the immunosuppressive tumor microenvironment and overcome PARPi resistance in ovarian cancer.

Summary: This study investigates the neuroprotective mechanism of CRLF3. By studying the expression and proapoptotic function of acetylcholinesterase in insect neurons, it is found that activation of CRLF3 can prevent the enhanced expression of acetylcholinesterase under apoptogenic conditions, thereby exerting neuroprotective effects.

Summary: This study investigates the role of TET-mediated DNA hydroxymethylation in bone formation using a novel selective TET enzyme inhibitor, BC339. The results show that inhibiting TET with BC339 suppresses osteoblast differentiation and functions through increasing the methylation level of the Sp7 promoter.

Summary: This study elucidated the molecular mechanisms of LILRB2-mediated inhibition of TREM2 signaling in microglia and demonstrated a novel approach to enhance microglial functions by blocking LILRB2-ligand interactions. The LILRB2 antagonist antibody rescued the inhibition of TREM2 signaling by LILRB2, suggesting a potential therapeutic strategy for improving microglial functions.

Summary: Homelessness has been on the rise in Europe, but there is a lack of comprehensive health data on this population. A study in Hungary found that homeless individuals have poorer health outcomes, higher rates of chronic diseases, lower healthcare utilization, and higher rates of smoking compared to the general population and the lowest income quintile. This calls for specific interventions to improve the health of homeless individuals.
