Summary: A protocol for inducing PLB development in Paphiopedilum SCBG Huihuang90 was established, revealing significant polarity and cell size gradients within each PLB. The increase in IAA and JA levels, along with decreased GA levels, indicated their correlation with PLB development. Transcriptomic profiles of different PLB developmental stages identified key genes involved in PLB development, with DEGs related to secondary metabolite metabolism and photosynthesis, as well as hormone signaling. This study provides valuable insights into the mechanisms of PLB development in Paphiopedilum.

Summary: This study found that the air-lift bioreactor had the best proliferation effect on Dendrobium loddigesii protocorm-like bodies (PLBs) compared to other culture methods. The optimal inoculum quantity was determined to be 30 g/vessel, cultured for 28 days in the bioreactor. Analysis of PLB extracts revealed that the ethyl acetate (EtOAc) extract had the highest levels of flavonoids and alkaloids, as well as potent antioxidant activity. Furthermore, the EtOAc extract showed significant antiproliferative effects on human colon carcinoma cells.

Summary: Protocorm-like bodies (PLBs) of Dendrobium officinale were obtained through tissue culture. The optimal medium formula for PLB propagation was determined. Stems, PLBs, and leaves of D. officinale had the highest content of different bioactive ingredients. Comparative transcriptome analysis revealed differential gene expression related to biosynthetic pathways of polysaccharides, alkaloids, and flavonoids in D. officinale.

Summary: This study characterized the morpho-anatomical aspects of germination and post-seminal development of C. crispa seeds, and investigated the effect of cryopreservation on these seeds. The results showed that cryopreservation can be used for long-term conservation of the seeds. These findings contribute to the overall understanding of the biology of C. crispa and the preservation of genetic material.

Summary: During fertilization and embryo development, the cellular totipotency undergoes changes, with some embryonic stem cells being able to convert to a totipotent state. In the process of somatic cell nuclear transfer (SCNT), however, the establishment of totipotency in cloned embryos is often compromised. This review explores practical strategies for inducing 2-cell-like cells (2CLCs) in the study of cloned embryo development and suggests that these cell models can offer new opportunities for improving SCNT efficiency.

Summary: Centaury, a medicinal plant with exceptional developmental plasticity, undergoes vigorous regeneration via shoot organogenesis and somatic embryogenesis, with arabinogalactan proteins (AGPs) playing a crucial role. Using machine learning prediction of proline hydroxylation, AGP sequences in centaury transcriptome were identified and analyzed. The study revealed AGPs with low glycomotif representation and discussed the expression profiles of selected AGP subclasses during various developmental stages and tissue samples.
