A free radical-generating system induces the cholesterol biosynthesis pathway: a role in Alzheimer's disease

Oxidative stress, which plays a critical role in the pathogenesis of neurodegenerative diseases such as Alzheimer's disease (AD), is intimately linked to aging - the best established risk factor for AD. Studies in neuronal cells subjected to oxidative stress, mimicking the situation in AD brains, are therefore of great interest. This paper reports that, in human neuronal cells, oxidative stress induced by the free radical-generating xanthine/xanthine oxidase (X-XOD) system leads to apoptotic cell death. Microarray analyses showed a potent activation of the cholesterol biosynthesis pathway following reductions in the cell cholesterol synthesis caused by the X-XOD treatment; furthermore, the apoptosis was reduced by inhibiting 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGCR) expression with an interfering RNA. The potential importance of this mechanism in AD was investigated by genetic association, and it was found that HMGCR, a key gene in cholesterol metabolism and among those most strongly upregulated, was associated with AD risk. In summary, this work presents a human cell model prepared to mimic the effect of oxidative stress in neurons that might be useful in clarifying the mechanism involved in free radical-induced neurodegeneration. Gene expression analysis followed by genetic association studies indicates a possible link among oxidative stress, cholesterol metabolism and AD.

Summary: In this article, the potential of machine learning in the differential diagnosis of systemic lupus erythematosus and primary Sjogren's syndrome using gene expression and methylation data from 651 individuals is demonstrated. The impact of disease heterogeneity on predictive model performance is analyzed, with patients assigned to specific molecular clusters being misclassified more frequently and affecting overall model performance. Additionally, biomarkers that improve predictions compared to using the entire dataset are identified and validated with external studies.

Summary: Mosaic loss of chromosome Y (mLOY) is a common somatic event associated with Alzheimer's disease (AD) and can be age-confounded in genome-wide association studies. This study used Mendelian randomisation to construct an age-independent mLOY polygenic risk score (mloy-PRS) and found that higher genetic risk for mLOY was associated with faster progression to AD in men with mild cognitive impairment. The study suggests that mLOY is involved in AD pathogenesis and shows associations with cerebrospinal fluid biomarkers.

Summary: We have developed compounds that show promising activity against antibiotic-resistant bacteria, Acinetobacter baumannii and Pseudomonas aeruginosa, which are on the WHO priority list. Compound 27, derived from DNA gyrase inhibitor 1, exhibited improved solubility, inhibition of topoisomerase IV, and no cross-resistance to novobiocin. Further optimization steps led to analogs of 27 with improved plasma distribution and retained anti-Gram-negative activity.

Antiparasitic Meroterpenoids Isolated from Memnoniella dichroa CF-080171

Summary: Memnoniella is a fungal genus that produces a variety of biologically active compounds. A specific extract from Memnoniella dichroa CF-080171 showed potent activity against Plasmodium falciparum 3D7 and Trypanosoma cruzi Tulahuen parasites. Eight new meroterpenoids with varying potencies were isolated from the extract, and compounds 1, 2, and 4 exhibited strong antiparasitic activity against the mentioned parasites and cytotoxic activity against HepG2 cells.

Summary: A new series of dual low nanomolar benzothiazole inhibitors of bacterial DNA gyrase and topoisomerase IV were developed, exhibiting broad-spectrum antibacterial activities against various strains. Cognitive decline in diabetic mice predisposed to Alzheimer?s disease is greater than in wild type

Summary: This study tested the hypothesis that individuals with type 2 diabetes (T2DM) will only develop dementia if they have a genetic predisposition to neurodegenerative disease. To support this concept, T2DM was induced in middle-aged hAPP NL/F mice, a preclinical model of Alzheimer's disease. The results showed that T2DM led to more severe behavioral, electrophysiological, and structural changes in these mice compared to wild-type mice. Mechanistically, the deficits were not caused by increased levels of toxic A beta or neuroinflammation, but by reduced gamma-secretase activity, decreased levels of synaptic proteins, and elevated tau phosphorylation. RNA-seq analysis suggested that hAPP NL/F mice may be more susceptible to T2DM due to defects in trans-membrane transport. These findings confirm the importance of genetic background in the severity of cognitive disorders in individuals with T2DM and suggest the inhibition of gamma-secretase activity as one of the underlying mechanisms.

Summary: Cancer, characterized by an alteration of the oxidative metabolism, is a major cause of human death worldwide. Many natural compounds from plant origin with anti-tumor attributes have been described, including capsaicin, the molecule responsible for the pungency in hot pepper fruits, which has shown anti-proliferative properties against cancer. The study found that the capsaicin content in pepper fruits did not correspond with their anti-proliferative activity, indicating the presence of other compounds with anti-tumor potentiality in pepper fruits.

Summary: This study found no significant relationship between the concentration of A beta(40) in the cerebrospinal fluid (CSF) and clinical symptoms or disease progression in Alzheimer's disease (AD) patients. However, A beta(40) was positively correlated with phosphorylated Tau (p-Tau) and total Tau concentrations, supporting their potential interaction in AD pathophysiology.

Summary: This study aimed to investigate the clinical indications, complications, and operational techniques of TPE treatment in patients with neurological diseases. The results showed that TPE treatment was well-tolerated by patients, with the main complications being hypotensive/vasovagal reactions. GBS patients were more prone to complications compared to MG patients.
