Magnetic affinity enzyme-linked immunoassay based on recombinant 26 kDa glutathione-S-transferase for serological diagnosis of schistosomiasis japonica

Schistosomiasis remains a serious worldwide public health problem. Improving the diagnostic assay for surveillance and monitoring will contribute to hastening the possible elimination of the disease in endemic regions. Therefore, this study aims to develop magnetic affinity enzyme-linked immunoassay (MEIA) for serological diagnosis of schistosomiasis based on recombinant 26 kDa glutathione-S-transferase of Schistosoma japonicum (rSj26GST). BALB/c mice infected with S. japonicum cercariae (40 per mouse) were used. After infecting for 6 weeks, the antibody was detected by MEIA. All of the infected mouse sera were effectively determined by MEIA. Compared with the enzyme-linked immunosorbent assay (ELISA). MEIA has a higher ratio of the mean positive value to the mean negative value (P/N) at the same dilution ratio (3.92 versus 2.66). MEIA was further applied for diagnosis of human schistosomiasis. Sera from 28 schistosomiasis-confirmed patients with low-intensity infection, 15 treated patients, and 20 non-endemic negative controls, were used to assess the assay. The results showed that MEIA and ELISA had similarity in positive detection rates. However, the higher P/N of MEIA was observed at the same dilution ratio. MEIA had high negative rate in detection of specific IgG in the treated patients. Moreover, there was no cross reaction with the sera of paragonimiasis patients. These results suggested that MEIA based on rSj26GST is a simple, rapid, convenient assay for the diagnosis of schistosomiasis. (C) 2012 Elsevier B.V. All rights reserved.

The immune-chemo-embolization effect of temperature sensitive gold nanomedicines against liver cancer

Radiofrequency-Activated Pyroptosis of Bi-Valent Gold Nanocluster for Cancer Immunotherapy

Summary: Pyroptosis of tumor cells was induced by biGC@PNA with a precisely stoichiometric ratio of Au(I) ion/Au(0) atom through its radiofrequency (RF)-heating effect. The activated pyroptosis resulted in a robust immunogenic cell death (ICD) effect, enhancing the antitumor immune responses and suppressing tumor metastasis and relapse. The noninvasive RF field provides a promising strategy for improving cancer immunotherapy.

Summary: This study developed a small molecular prodrug-based nanomedicine with high drug-loading efficiency and tumor selectivity for the treatment of solid tumors, especially triple negative breast cancers. The nanomedicine actively targeted TNBC 4T1 tumor tissues and achieved reduction-responsive drug release. It significantly suppressed tumor growth and eliminated cancer stem cells by disrupting the tumor microenvironment and consuming intracellular glutathione. This research provides valuable insights for the rational design of prodrug nanomedicines for the treatment of stroma- and CSCs-rich solid malignancies.

Targeting cancer-associated fibroblasts with hydroxyethyl starch nanomedicine boosts cancer therapy

Summary: Researchers designed a nanomedicine that specifically targets cancer-associated fibroblasts (CAFs) in triple-negative breast cancer (TNBC) by conjugating doxorubicin-loaded hydroxyethyl starch-IR780 nanoparticles with a CREKA peptide. The nanomedicine effectively targeted CAFs and generated hyperthermia upon light irradiation, leading to a reduction in CAFs through chemo- and photothermal-therapies. By modulating the tumor mechanical microenvironment (TMME) and eliminating cancer stem cells (CSCs), the growth of the 4T1 tumor was significantly inhibited. This strategy provides a novel therapeutic treatment for desmoplastic solid tumors.

Mild photothermal therapy boosts nanomedicine antitumor efficacy by disrupting DNA mechanics damage repair pathways and modulating tumor

Summary: Various PARPi have been approved for the treatment of breast cancers, but they are only effective for HR-deficient breast cancers. The abnormal micro-environment of breast cancers hinders drug delivery. This study used mesoporous polydopamine to deliver PARPi and a DNA-damaging drug, and demonstrated that local mild photothermal therapy could inhibit the HR repair pathway and repress cancer-associated fibroblasts, thereby enhancing the antitumor effect and improving drug delivery.

A novel lonidamine derivative targeting mitochondria to eliminate cancer stem cells by blocking glutamine metabolism

Summary: In this study, a novel derivative of lonidamine (LND) called HYL001 was found to selectively and potently inhibit cancer stem cells (CSCs) by targeting mitochondria, with significantly lower IC50 values compared to LND. Mechanistically, HYL001 downregulates glutaminase expression to block glutamine metabolism, leading to apoptotic cell death. HYL001 also displays significant antitumor activity in vivo and represses CSCs in fresh tumor tissues from liver cancer patients.

Mechano-boosting nanomedicine antitumour efficacy by blocking the reticuloendothelial system with stiff nanogels

Summary: The authors designed a series of nanogels with different stiffness and investigated how the mechanical properties of the nanogels can overcome the limitations of RES. Stiff nanogels were able to block uptake capacity of macrophages and temporarily block RES, while soft nanogels efficiently delivered drugs. Combining the advantages of stiff and soft nanogels led to the optimum tumor inhibition effect.

Prevalence of zoonotic (brugian) filariasis in Asia: A proportional meta-analysis

Looking through the lens of social science approaches: A scoping review of leishmaniases and Chagas disease research

Rapid, sensitive, and convenient detection of Plasmodium falciparum infection based on CRISPR and its application in detection of asymptomatic infection

Summary: Rapid and convenient detection of Plasmodium falciparum, the deadliest malaria parasite in Africa, is crucial for malaria eradication efforts in medical resource-limited settings. A CRISPR-based diagnostic method that can detect P. falciparum without nucleic acid extraction was developed and showed high sensitivity and consistency with qPCR test. It can also detect artemisinin-resistant strains and other laboratory-cultured strains. This method has great potential for clinical testing and large-scale community screening in Africa and other regions.

High detection frequency and genetic diversity of porcine circovirus 3 (PCV-3) in Namibian backyard farms and warthogs

Summary: Porcine circovirus 3 (PCV-3) has been reported globally since its first identification in 2015. Limited data on PCV-3 in Africa led to a study in Namibia, which found a high prevalence of PCV-3 in backyard piggeries, but not in commercial farms with higher biosecurity measures. The source of viral introduction and factors such as semen importation, feeding backyard pigs with human food waste, pig exchanges, and wildlife movements may be involved in the dispersal of PCV-3 within Namibia. Further studies are needed to understand the epidemiology and impact of PCV-3 on pig productivity and wildlife health.

Natural SARS-CoV-2 infection in dogs: Determination of viral loads, distributions, localizations, and pathology

Biomarker of Anopheles exposure in Ghanaian children with hemoglobin S and C

Summary: Heterozygous carriers of HbAS and HbAC have a reduced risk of severe malaria, but not immunity against Plasmodium falciparum infection. This study highlights the importance of studying the role of hemoglobinopathies in malaria transmission for improving control interventions.

Spatial analysis of human and livestock anthrax in Lai Chau province, Vietnam (2004-2021)

Spatial clusters of human and livestock anthrax were identified, and the characteristics of the disease were described. The study found that there were distinct differences between the high-risk areas for human and livestock anthrax, and most human cases occurred during specific seasons. Additionally, the decrease in livestock anthrax vaccine coverage was associated with an increase in disease incidence.
