Summary: Multiple sclerosis (MS) is a chronic immune-mediated disease with no known treatment yet. Mesenchymal stem cells (MSCs) show promise in MS treatment by their ability to migrate and home into the CNS. The chemokine/chemokine receptor axes play a pivotal role in MSC trafficking and their targeted tropism to the CNS in MS patients.

Summary: Pro-inflammatory CD4(+) T helper cells play a crucial role in autoimmune diseases. Recent research has revealed the heterogeneity of chemokine receptors expressed by these cells, which are responsible for recruiting different subsets of pro-inflammatory Th cells in autoimmune inflammation. Understanding these receptors may lead to potential therapeutic targets for intervention.

Summary: Cell therapies offer hope in treating neurodegenerative diseases and spinal cord injuries by promoting functional neural circuit reconstruction and slowing disease progression. Comprehensive characterization of cells before transplantation is crucial for safety, as it can prevent risks such as tumorous growth due to undifferentiated cell proliferation.

Summary: This study aimed to investigate the characteristics and distribution of HIV-infected cells in semen, in order to improve strategies for exposure prevention and assisted reproduction for HIV-infected partners. The results showed that HIV-infected spermatozoa were mainly CD4 negative, and p24-positive cells in semen were predominantly spermatozoa. Additionally, the study confirmed that motile spermatozoa could carry HIV into peripheral blood mononuclear cells of healthy men.

Summary: Gaucher disease is a lysosomal storage disease caused by mutations in GBA1/Gba1. Deficiency of lysosomal acid beta-glucosidase enzyme leads to abnormal accumulation of glucosylceramide, resulting in altered function of immune cells and tissue damage. The study reveals the role of CXCR3 receptor and CXCL9 chemokine in increased recruitment of T cells in Gaucher disease, suggesting potential therapeutic targets for inflammation treatment.

Summary: Multipotent mesenchymal stromal cells (MSCs) are promising cellular therapeutics for treating inflammatory and degenerative disorders due to their anti-inflammatory, immunomodulatory, and regenerative potentials. MSCs can be obtained from various tissues, with bone marrow being the most commonly used starting material for clinical applications. Understanding the expression of chemokine receptors and ligands by MSCs from different tissues is crucial for predicting their therapeutic potential.

Summary: This study investigated the effects of erythropoietin (EPO) on the differentiation of neural stem cells (NSCs)/neural progenitors (NPs) in hypoxic-ischemic injury. Results showed that EPO promoted the differentiation of NSCs/NPs into oligodendrocytes and astrocytes, while inhibiting neuronal differentiation. The EPO receptor/β common receptor heterodimer was found to be present on the surface of fetal NSCs/NPs and involved in glial differentiation. The study provides insights into the potential mechanisms of EPO in treating hypoxic-ischemic injury.

Summary: Experimental autoimmune encephalomyelitis (EAE) is an animal model used to mimic multiple sclerosis (MS), with the innate immune response playing a key role in both acute and chronic stages of MS. The composition and roles of microglia, CNS resident myeloid units, and infiltrating myeloid cells differ during EAE, with specific myeloid cell types and states being responsible for antigen presentation and disease propagation. Single-cell techniques offer insight into proinflammatory and beneficial cell states, as well as identification of potential treatment targets for EAE.

Summary: Baicalein (BAI), a natural flavonoid, has been found to alleviate clinical symptoms, demyelination, and inflammatory response in EAE mice. It inhibits M1 microglia/macrophage activation, decreases the expression of proinflammatory cytokines, and targets STAT1 to inhibit polarization. These findings suggest the potential use of BAI in the treatment of MS.

Summary: In multiple sclerosis, pathogenic T cell responses play a major role in autoimmune inflammation, but B cells may also contribute to the pathogenesis through antigen presentation and the production of proinflammatory cytokines. This study developed a new model to study the roles of pathogenic B cells in the development of multiple sclerosis, providing insights into the disease's pathogenic processes.
