Schiff bases, an aldehyde-or ketone-like compounds in which the carbonyl group is replaced by an imine or azomethine, are some of the most widely used organic compounds. Indeed, they are widely used for industrial purposes and also exhibit a broad range of biological activities, including anti-urease activity. Ureases, enzymes that catalyze urea hydrolysis, have received considerable attention for their impact on living organisms' health, since the persistence of urease activity in human and animal cells can be the cause of some diseases and pathogen infections. This short review compiles examples of the most anti-urease Schiff bases (0.23 mu M < IC50 < 37.00 mu M) and their metal complexes (0.03 mu M < IC50 < 100 mu M). Emphasis is given to ureases of Helicobacter pylori and Canavalia ensiformis, although the active site of this class of hydrolases is conserved among living organisms. Summary: The study introduces two novel calix[n]arene-based anti-cocaine immunogens (V4N2 and V8N2) and evaluates their preclinical efficacy. The immunogens induce high titers of cocaine antibodies and modify the biodistribution of a cocaine analogue, reducing cocaine-induced behavior. These findings demonstrate the potential of V4N2 and V8N2 as immunogens for the treatment of cocaine use disorder.

Anticancer properties of arylchromenes and arylchromans: an overview

Interaction and inhibition mechanism of urease in vitro and soil system by a natural benzylisothiocyanate isolated from Moringa oleifera

Summary: Urease catalyzes urea hydrolysis in the soil, leading to N-urea losses by volatilization. This study evaluated the mechanism of urease inhibition by MFC, a compound isolated from Moringa oleifera L. The results showed that MFC is a mixed inhibitor and interacts with urease, causing conformational changes in the protein structure. The interaction also leads to the formation of dithiocarbamates and thiourea, which are inhibitors of urease. MFC shows potential as a biotechnological application compound.

Thiohydantoins and hydantoins derived from amino acids as potent urease inhibitors: Inhibitory activity and ligand-target interactions

Summary: This study investigated the inhibitory effect of hydantoins and thiohydantoins derived from L and n-amino acids on Canavalia ensiformis urease (CEU). The results showed that most thiohydantoins exhibited promising inhibitory potential, with six compounds showing higher inhibitory activity (%I) than the reference inhibitor thiourea (56.5%). Among them, thiohydantoin derived from L-valine (1b) and hydantoin derived from L-methionine (2d) were identified as the most potent inhibitors. The kinetic studies revealed that these compounds exhibited mixed and uncompetitive inhibition, with defined Ki values. Molecular docking studies suggested that the thiohydantoin and hydantoin rings can act as a pharmacophoric group, forming hydrogen bonding interactions with critical amino acid residues in the enzyme active or allosteric site. The stereochemistry of the compounds also influenced their inhibition, kinetics, and binding parameters.

Bis-benzimidazolium salts as bifunctional organocatalysts for the cycloaddition of CO2 with epoxides

Summary: A series of 1,2-bis(benzimidazolium)ethane dihydrohalide salts were synthesized and characterized for the cycloaddition reaction of CO2 with epoxides. The salts containing 1,2-bis(5-methyl-benzimidazolium)ethane cation showed the best activity, and the study of various parameters confirmed the stability and high selectivity of the catalyst. A catalytic mechanism was proposed based on H-1 NMR spectroscopy experiments. The simple, nontoxic, and low-cost synthesis of these catalysts, along with high conversions at low catalyst loading, makes them promising for the production of cyclic carbonates using carbon dioxide.

Summary: In this study, a novel H2S-releasing phthalimide hybrid was synthesized and demonstrated to have activity in models of inflammatory pain. The compound exerted its effects through the release of H2S and activation of ATP-sensitive potassium channels. This finding stimulates further research to evaluate H2S-releasing phthalimide hybrids as candidate analgesic drugs.

Inclusion complex of ketoconazole and p-sulfonic acid calix[6]arene improves antileishmanial activity and selectivity against Leishmania amazonensis and Leishmania infantum

Summary: In this study, an inclusion complex (IC6HKTZ) of ketoconazole (KTZ) with p-sulfonic acid calix[6]arene (CX6SO3H) was prepared to improve the solubility and efficacy of KTZ against Leishmania amazonensis and Leishmania infantum promastigotes. The solubility enhancement of KTZ by 120.00 mu M CX6SO3H was about 95 times. IC6HKTZ exhibited lower IC50 values compared to free KTZ for both Leishmania species, indicating improved anti-leishmanial activities.

Safety and immunogenicity of the anti-cocaine vaccine UFMG-VAC-V4N2 in a non-human primate model

Summary: An effective method for treating cocaine addiction is the use of anti-drug vaccines. These vaccines stimulate the production of antibodies that can bind to cocaine and reduce its passage through the blood-brain barrier, thereby decreasing its activity in the brain. A new vaccine candidate, UFMG-V4N2, was developed by our research group using an innovative carrier based on calixarenes. Safety and immunogenicity of the anti-cocaine vaccine were assessed in non-human primates, and it was found that UFMG-VAC-V4N2 had minimal side effects and did not affect general health or other physiological parameters. This preclinical study provides important evidence for future clinical trials of the anti-cocaine vaccine.

Summary: Twenty chromenes were synthesized with good to excellent yields (70-96%). Compounds 14, 12, 15, 16, 17, 18, and 20 showed the strongest inhibitory effects on human cancer cell lines. The presence of para-position electron-withdrawing substituents on the phenyl ring favored potency and selectivity for cancer cells. Biophysical studies indicated that certain chromene derivatives preferentially interacted with ctDNA via intercalation, which was consistent with the in vitro biological results.

Summary: The toxicological effects of 25H-NBOMe and 25H-NBOH on zebrafish embryos and larvae were demonstrated, including both lethal and non-lethal effects. These drugs not only caused high embryo mortality, but also various malformations and adverse effects. In vitro experiments showed that the drugs have an affinity for DNA, leading to conformational changes in the DNA structure.
