Stem cells derived from human dental pulp tissue (DP-MSC) differ from the other mesenchymal stem cells prepared from bone marrow or adipose tissue due to their embryonic origin from the neural crest and are of special interest because of their neurotropic character. Furthermore, the therapeutic potential of DP-MSCs is realized through paracrine action of extracellularly released components, for which exosomes play an important role. In this review, we intend to explore the properties of these cells with an emphasis on exosomes. The therapeutic applicability of these cells and exosomes in dental practice, neurodegenerative diseases, and many other difficultly treatable diseases, like myocardial infarction, focal cerebral ischemia, acute lung or brain injury, acute respiratory distress syndrome, acute inflammation, and several others is concisely covered. The use of cellular exosomes as an important diagnostic marker and indicator of targeted cancer therapies is also discussed, while the importance of stem cells from human exfoliated deciduous teeth as a source of evolutionally young cells for future regenerative therapies is stressed. We conclude that exosomes derived from these cells are potent therapeutic tools for regenerative medicine in the near future as clinical administration of DP-MSC-conditioned medium and/or exosomes is safer and more practical than stem cells.

Intracellular prodrug gene therapy for cancer mediated by tumor cell suicide gene exosomes

Ursula Altanerova, Jana Jakubechova, Katarina Benejova, Petra Priscakova, Vanda Repiska, Andrea Babelova, Bozena Smolkova, Cestmir Altaner

Summary: Exosomes containing suicide gene mRNA secreted from engineered MSCs have the potential to be a specific therapy for tumors and potentially for metastases by inducing cell death in tumor cells without inhibiting the growth of normal cells. This innovative approach utilizes exosomes as a delivery system for suicide genes, showing promising results in cancer treatment.

Selective Apoptotic Effect of Plasma Activated Liquids on Human Cancer Cell Lines

Summary: Plasma medicine is a new field focusing on biomedical and clinical applications of cold gas plasmas, including their anticancer effects. Effects of plasma-activated liquids (PAL) on cancer cells showed selectivity compared to normal fibroblasts, depending on correctly chosen activation time and PAL concentration. Plasma-generated hydrogen peroxide may play a role in the selectivity effect of PAL on cancer cells.

Summary: This study investigated the production of peptaibols by Trichoderma atroviride O1 and their antibacterial and anticancer activity against drug-sensitive and multi-drug resistant bacteria and cancer cell lines. The results showed that peptaibols exhibited significant inhibitory effects on the growth of methicillin-resistant Staphylococcus aureus and human breast cancer and ovarian cancer cell lines.

Summary: Extracellular vesicles secreted by human mesenchymal stem/stromal cells can be used to treat high-grade glioblastomas. These therapeutic vesicles, engineered with a gene, can inhibit the growth of glioblastoma cells in the presence of a specific drug. Animal experiments have shown the potential of this new cell-free therapy for tumor treatment.

Liquid Biopsy as a Source of Nucleic Acid Biomarkers in the Diagnosis and Management of Lynch Syndrome

Gergely Buglyo, Jakub Styk, Ondrej Pos, Adam Csok, Vanda Repiska, Beata Soltesz, Tomas Szemes, Balint Nagy

Summary: Lynch syndrome is an inherited cancer predisposition disorder that can lead to various types of cancers. Current diagnosis methods involve testing for MMR protein deficiency, mutation detection, microsatellite instability, and MLH1 promoter hypermethylation. Recent studies have shown the potential of using DNA and RNA markers extracted from liquid biopsies for diagnosing LS.

Summary: This article provides an overview of the application of liquid biopsy in the diagnosis and management of colorectal cancer (CRC). It discusses the use of liquid biopsy-derived DNA and RNA biomarkers for early detection and monitoring of disease recurrence. The article also explores the genetic causes of CRC, the advantages and difficulties of liquid biopsy, and the potential for personalized management.

Extracellular vesicles derived from dental mesenchymal stem/stromal cells with gemcitabine as a cargo have an inhibitory effect on the growth of pancreatic carcinoma cell lines in vitro

Daniela Klimova, Jana Jakubechova, Ursula Altanerova, Andreas Nicodemou, Jakub Styk, Tomas Szemes, Vanda Repiska, Cestmir Altaner

Summary: Extracellular vesicles (EVs) are utilized as a drug delivery tool in cancer therapy. We engineered mesenchymal stem/stromal cells (MSCs) to secrete EVs packed with chemotherapeutic agents, which were then delivered directly into tumor cells. The EVs inhibited the growth of pancreatic carcinoma cell lines in vitro, and the MSCs were also engineered to express a suicide gene that converted a non-toxic prodrug into a highly cytotoxic drug in cancer cells, further inhibiting their growth.
