Summary: Novel indole-piperazine derivatives with a hydroxamic acid moiety were synthesized as selective HDAC6 inhibitors, showing nanomolar IC50 values in enzymatic assays. Among them, compound 9c exhibited the highest potency with an IC50 of 13.6 nM. In vitro studies showed that 9c induced neurite outgrowth and had neuroprotective effects against oxidative damage in PC12 cells, surpassing Tubastatin A. Western blot assay demonstrated that 9c increased the acetylation of alpha-tubulin in a dose-dependent manner.

Summary: The neural cell adhesion molecule L1 plays important roles in cell migration, survival, differentiation, myelination, neurite outgrowth, and signaling in nervous system development and in adults. The proteolytic cleavage of L1 generates soluble and transmembrane fragments that regulate cellular functions. The study identifies the interaction between the extracellular LIR motif of L1 and LC3, which is important for neuronal survival and neurite outgrowth.

Summary: RAB35 is a multifunctional small GTPase that regulates endocytic recycling, cytoskeletal rearrangement, and cytokinesis. Our findings revealed that RAB35 is required for precise neuronal distribution in the developing hippocampus by regulating the expression of cell adhesion molecules, thereby influencing spatial memory. Mice with a brain-specific knockout of the Rab35 GTPase exhibit defects in spatial memory and hippocampal lamination, outlining a key role for Rab35 in hippocampal development and function.

Summary: This study focused on the neurotrophic activity of derivatives based on two natural sesquiterpene compounds. Most of the derivatives showed stronger neurite outgrowth-promoting activity compared to the original compounds, merrillianone and cycloparviforalone. Compounds 2a, 3a, and 3b exhibited the most potent neurite outgrowth-promoting activity among all target derivatives.

Summary: The loss of structure and function of neurons, along with cognitive decline, is commonly observed in neurologic diseases. This study reveals that increased expression of MBNL2 is associated with neuronal maturation and is essential for neuronal morphogenesis and the transition from fetal to adult RNA processing. The findings suggest that calpain-2-mediated MBNL2 degradation and re-induction of developmental RNA processing may contribute to neurodegeneration.
