Inhibition of KRAS-dependent lung cancer cell growth by deltarasin: blockage of autophagy increases its cytotoxicity

Deltarasin is a recently identified small molecule that can inhibit KRAS-PDEd interactions by binding to a hydrophobic pocket on PDEd, resulting in the impairment of cell growth, KRAS activity, and RAS/RAF signaling in human pancreatic ductal adenocarcinoma cell lines. Since KRAS mutations are the most common oncogene mutations in lung adenocarcinomas, implicated in over 30% of all lung cancer cases, we examined the ability of deltarasin to inhibit KRAS-dependent lung cancer cell growth. Here, for the first time, we document that deltarasin produces both apoptosis and autophagy in KRAS-dependent lung cancer cells in vitro and inhibits lung tumor growth in vivo. Deltarasin induces apoptosis by inhibiting the interaction of with PDEd and its downstream signaling pathways, while it induces autophagy through the AMPK-mTOR signaling pathway. Importantly, the autophagy inhibitor, 3-methyl adenine (3-MA) markedly enhances deltarasin-induced apoptosis via elevation of reactive oxygen species (ROS). In contrast, inhibition of ROS by N-acetylcysteine (NAC) significantly attenuated deltarasin-induced cell death. Collectively, these observations suggest that the anti-cancer cell activity of deltarasin can be enhanced by simultaneously blocking tumor protective autophagy, but inhibited if combined with an anti-oxidant.

Summary: This study successfully synthesized hydrophobic mesoporous poly(ionic liquid)s (PILs) with strong Bronsted-Lewis acid sites and demonstrated their efficiency as catalysts for the selective valorization of lignite into monocyclic aromatic compounds (MCACs) with a yield of 23.5 wt.%. The synergistic catalysis mechanisms include the hydrophobic micro-environment created by the PILs, the mesoporous confinement that enhances substrate concentration, and the active hydrogen transfer of the Bronsted-Lewis acid sites.

The role of non-coding RNAs (miRNA and lncRNA) in the clinical management of rheumatoid arthritis

Summary: Rheumatoid arthritis is a chronic systemic inflammatory disorder associated with autoimmune response dysregulation. Recent advancements in early diagnosis, aggressive treatment and alternative therapeutic options have improved the management and long-term prognosis of the disease. Non-coding RNA, such as microRNA and long non-coding RNA, has been found to be deregulated in various diseases, including rheumatoid arthritis, and may contribute to its pathogenesis, disease progression and treatment. This review summarizes the changes in non-coding RNAs and their underlying mechanisms in the development and progression of rheumatoid arthritis, and highlights novel therapeutic targets/strategies and biomarkers for its therapy.

Bienzymatic Cascade Combining a Peroxygenase with an Oxidase for the Synthesis of Aromatic Aldehydes from Benzyl Alcohols

Summary: This study proposes a parallel cascade using aryl alcohol oxidase (PeAAOx) from Pleurotus eryngii and unspecific peroxygenase (AaeUPO) from Agrocybe aegerita to convert aromatic primary alcohols into high-value aromatic aldehydes. Key factors such as enzyme dosage, pH, and temperature were investigated. The study evaluates the universality of PeAAOx coupled with AaeUPO cascade catalysis for the synthesis of aromatic aldehyde flavor compounds.

Suppression of PD-L1 release from small extracellular vesicles promotes systemic anti-tumor immunity by targeting ORAI1 calcium channels

Summary: Blockade of immune checkpoints in cancer cells through the expression of PD-L1 has received significant attention. In this study, we investigate the role of Ca2+-dependent proteins in regulating release of PD-L1-containing small extracellular vesicles (sEVs) and tumor growth in mouse models. Our findings suggest that sEVs release in cancer cells is Ca2+-dependent, and knockdown of the ORAI1 gene reduces Ca2+-signals and sEVs release, reinvigorating T cell response and slowing tumor progression. Additionally, we identify ORAI1 upregulation as an unfavorable prognostic factor for non-small-cell lung cancer patients.

Summary: In this study, pomiferin, a natural product from Osage Orange, was found to activate autophagy and inhibit drug efflux to overcome innate and acquired drug resistance in cancer cells. It also exhibited tumor suppression and anti-metastatic effects in animal models. Pomiferin could serve as a novel effective agent for circumventing drug resistance in clinical applications.

This review discusses the effects of extracorporeal shock waves, low-intensity pulsed ultrasound, and mechanical stress on tendon-bone healing, focusing on their possible mechanisms of action. The aim is to provide potential therapeutic approaches for clinical treatment.
