Pathway based therapeutic targets identification and development of an interactive database CampyNIBase of Campylobacter jejuni RM1221 through non-redundant protein dataset

The bacterial species Campylobacter jejuni RM1221 (CjR) is the primary cause of campylobacteriosis which poses a global threat for human health. Over the years the efficacy of antibiotic treatment is becoming more fruitless due to the development of multiple drug resistant strains. Therefore, identification of new drug targets is a valuable tool for the development of new treatments for affected patients and can be obtained by targeting essential protein(s) of CjR. We conducted this in silico study in order to identify therapeutic targets by subtractive CjR proteome analysis. The most important proteins of the CjR proteome, which includes chokepoint enzymes, plasmid, virulence and antibiotic resistant proteins were annotated and subjected to subtractive analyses to filter out the CjR essential proteins from duplicate or human homologous proteins. Through the subtractive and characterization analysis we have identified 38 eligible therapeutic targets including 1 potential vaccine target. Also, 12 potential targets were found in interactive network, 5 targets to be dealt with FDA approved drugs and one pathway as potential pathway based drug target. In addition, a comprehensive database 'CampyNIBase' has also been developed. Besides the results of this study, the database is enriched with other information such as 3D models of the identified targets, experimental structures and Expressed Sequence Tag (EST) sequences. This study, including the database might be exploited for future research and the identification of effective therapeutics against campylobacteriosis.

Summary: This study aimed to investigate the genetic variants of kappa-casein and beta-lactoglobulin genes in native cattle. Genotyping analysis revealed three genotypes (AA, AB, and BB) for both genes, with the AA genotype being the most common for kappa-casein and the BB genotype being the most common for beta-lactoglobulin. The results indicated a high genetic diversity, but a deviation from Hardy-Weinberg equilibrium, suggesting the presence of selection pressure in the population.

Effect of gamma radiation on microbial load, physico-chemical and sensory characteristics of common spices for storage

Results showed that gamma radiation significantly reduced microbial load in spices without affecting their physico-chemical, nutritional, and sensory properties.

Wave-wise comparative genomic study for revealing the complete scenario and dynamic nature of COVID-19 pandemic in Bangladesh

Summary: This study analyzed over 1500 SARS-CoV-2 genome sequences from Bangladesh and found that the second wave of the pandemic had a significantly higher mutation rate and more deletion and transversion events compared to the first wave. Different virus lineages were predominant in different waves in Bangladesh, with the B.1.617 variant showing potential for increased transmissibility. Additionally, the study noted the absence of indigenous variants so far in the country, suggesting that preventing the import of foreign variants could help control the outbreak.

Engineering Nicotiana tabacum trichomes for triterpenic acid production

Summary: This study aimed to implement the biosynthesis of triterpenic acids in Nicotiana tabacum glandular trichomes by introducing five transgenes coding for key enzymes involved in the biosynthetic pathway. Analysis of metabolites extracted from entire leaves showed a low amount of ursolic acid in plants expressing the transgenes. These findings suggest that redirecting the terpene biosynthetic pathway in glandular trichomes of N. tabacum can be achieved without detrimental effects on the whole plant phenotype.

Molecular identification and high fidelity micropropagation of shell ginger (Alpinia zerumbet)

Summary: This study aimed to establish the DNA-based identity and a protocol for high-fidelity micropropagation of Alpinia zerumbet to support the medicinal plant industry. DNA barcode of A. zerumbet was successfully reported through PCR amplification and sequencing of rbcL and matK genes. Microshoots induced from rhizome buds on MS medium supplemented with 6.0 mg L-1 6-benzyl amino purine and 30 g L-1 sucrose rooted well in MS medium amended with 1.5 mg L-1 naphthalene acetic acid. The genetic uniformity of the micropropagated plants was confirmed through the analysis of random amplified polymorphic DNA and inter-simple sequence repeat primers. This research provides an accurate identification method for A. zerumbet and a micropropagation protocol for its economically feasible and environmentally sustainable exploitation.

Summary: This study analyzed the genomic diversity and phylogenetic relations of four dengue virus serotypes found in different regions, revealing close relationships between DENV3 and DENV1 strains, and distinct relationships with the other two strains (DENV2 and DENV4). DENV-2 serotype is considered more virulent with more epidemics of dengue hemorrhagic fever associated. PPIs between dengue virus proteins and its hosts in Bangladesh regions showed that dengue-human PPIs are more prevalent in DENV2 and DENV3 serotypes, while dengue-mosquito PPIs are most predominant in DENV1 serotype.
