Summary: This study revealed that the bud mutant RM from Red Delicious apple had significantly higher total anthocyanin content compared to the original strain OS. Metabolomics analysis found that cyanidin-3-O-galactoside was significantly accumulated in the fruit skin of RM. Transcriptome analysis showed up-regulation of the structural gene MdF3H and transcription factor MdMYB66 in the mutant. Overexpression of MdMYB66 promoted anthocyanin accumulation and increased the expression of related structural genes. The findings suggest that MdMYB66 regulates anthocyanin metabolism by precise gene expression regulation, providing insight into the biosynthesis and regulation of anthocyanins.
