BackgroundPlatelet-derived growth factor alpha (PDGFRA) is a gene encoding tyrosine kinase receptor and both EGFR and PDGFRA activate tyrosine kinases. The implication of PGFRA in many cancers and its prognostic significance irrespective to EGFR status in spinal chordoma, gliomas, and uterine cancers have shown a need for its investigation in oral squamous cell carcinoma (OSCC). We investigated the prognostic value of PDGFRA mRNA expression in OSCC. Patients and methodsThe study was conducted in the department of oral maxillofacial surgery-head and neck oncology, at a tertiary hospital. The data on PDGFRA mRNA expression and immunohistochemical staining status in primary OSCC patients treated for curative surgery from 2010 to 2012 were analyzed. Univariate and multivariate analyses were performed with other cofactors for survival. ResultsA total of 114 consecutive patients with primary OSCC who received treatment were studied. Thirty-one patients died of the disease. Strong PDGFRA immunohistochemical staining and high expression of PDGFRA mRNA were associated with positive pN status (P<.001), disease-free survival (P<.001), and overall survival (P<.001) in multivariate cox regression when all other factors such as pN status and histological grading were analyzed. Kaplan-Meier analysis revealed that the 2-year survival and 3-year survival of patients with PDGFRA mRNA low expression were 96.83%. However, 2-year survival for PDGFRA mRNA high expression level was 59.64%, which decreased to 45.57% by 3-years. ConclusionPDGFRA overexpression in oral SCC, in respect to strong PDGFRA immunohistochemical staining and high PDGFRA mRNA expression, was positively associated with regional metastasis and reduced patient survival.

Summary: This study analyzed the risk factors for survival in elderly patients with oral squamous cell carcinoma and developed a novel clinical signature for prognosis. Results indicated that high N stage, low hemoglobin level, low BMI, and high NLR were associated with poor survival outcomes. The constructed nomogram showed a good predictive ability.

Summary: The study revealed that high expression of CCL22 in T/MF SCC may promote the recruitment of Th2 cells and help predict a better survival. Additionally, mutations in NOTCH1 inhibit the differentiation of Th2 cells, facilitating tumor progression.

Summary: This study provides further insights into the risk factors for esophageal squamous cell carcinoma (ESCC) in patients with a history of head and neck squamous cell carcinoma (HNSCC). The study found that HNSCC patients, especially drinkers with an alcohol flushing response and those with high Ki67 labeling index (Ki67 LI) in HNSCC tissue, were more likely to develop ESCC.

Cancer cells co-opt nociceptive nerves to thrive in nutrient-poor environments and upon nutrient-starvation therapies

Yu Zhang, Chengzhong Lin, Zheqi Liu, Yiting Sun, Mingtao Chen, Yibo Guo, Wei Liu, Chenping Zhang, Wantao Chen, Jian Sun, Ronghui Xia, Yuhua Hu, Xi Yang, Jiang Li, Zhiyuan Zhang, Wei Cao, Shuyang Sun, Xu Wang, Tong Ji

Summary: This study reveals that cancer cells can utilize nociceptive nerves to survive in nutrient-poor environments. Cancer cells secrete NGF through ROS-triggered activation of c-Jun, which stimulates nerves to produce CGRP. The neurogenic CGRP induces cytoprotective autophagy in cancer cells through Rap1-mediated disruption of the mTOR-Raptor interaction. Blocking CGRP can inhibit the vicious cycle between cancer cells and nerves and therapeutically benefit nutrient-starvation therapies.

EVI2B Is a Prognostic Biomarker and Is Correlated with Monocyte and Macrophage Infiltration in Osteosarcoma Based on an Integrative Analysis

Zhenlin Dai, Zheqi Liu, Rong Yang, Wei Cao, Tong Ji

Summary: Osteosarcoma (OS), the most common malignant bone tumor, lacks updated treatment strategies. This study identified EVI2B as a significantly upregulated gene in OS tumor samples and demonstrated its association with chemokine signaling, cytokine-cytokine receptor interaction, and Human T-cell leukemia virus 1 infection. The prognostic prediction model including EVI2B, DOCK2, and CD33 indicated EVI2B as an independent protective prognostic marker in OS. The study also revealed the close relationship between EVI2B expression, monocyte and macrophage infiltration in OS, providing clear evidence for its prognostic value.

Yin Yang 1-Induced Long Noncoding RNA DUXAP9 Drives the Progression of Oral Squamous Cell Carcinoma by Blocking CDK1-Mediated EZH2 Degradation

Wenkai Zhou, Yisheng Feng, Chengzhong Lin, Chi Kuan Chao, Ziqi He, Shiyao Zhao, Jieyuan Xue, Xu-Yun Zhao, Wei Cao

Summary: A highly expressed lncRNA DUXAP9, found in oral squamous cell carcinoma (OSCC), is positively associated with poor prognosis. DUXAP9 promotes OSCC cell proliferation, migration, invasion, and tumor growth through the upregulation of N-cadherin, Vimentin, Ki67, PCNA, and EZH2 and downregulation of E-cadherin. Yin Yang 1 (YY1) activates the transcription of DUXAP9 in OSCC. Moreover, DUXAP9 interacts with EZH2 and inhibits its degradation, thereby blocking its translocation from the nucleus to the cytoplasm. DUXAP9 may serve as a promising therapeutic target for OSCC.

Characteristics of esophageal cancer in patients with head and neck squamous cell carcinoma

Lei Wang, Wenjing Pang, Kun Zhou, Lei Li, Feng Wang, Wei Cao, Xiangjun Meng

Summary: This study investigated the clinicopathological features of esophageal cancer in patients with a history of head and neck squamous cell carcinoma (HNSCC). Most cases were male patients aged 50-69 years, with higher proportion of early esophageal cancer and successful surgical treatment in the synchronous carcinoma group compared to the metachronous carcinoma group. Detection of esophageal cancer should be prioritized in HNSCC patients.
