Ethnopharmacological relevance: Vachellia (Acacia) nilotica and other plants of this genus have been used in traditional medicine of Asian and African countries to treat many disorders, including sexually transmitted diseases, but few studies were performed to validate their anti-microbial and anti-viral activity against sexually transmitted infections. Aim of the study: The present study was undertaken to explore whether the ethnomedical use of V.nilotica to treat genital lesions is substantiated by its antiviral activity against the human immunodeficiency virus (HIV), the herpes simplex virus (HSV) and the human papillomavirus (HPV). Materials and methods: The antiviral activity of V.nilotica was tested in vitro by virus-specific inhibition assays using HSV-2 strains, sensible or resistant to acyclovir, HIV-1IIIb strain and HPV-16 pseudovirion (PsV). The potential mode of action of extract against HSV-2 and HPV-16 was further investigated by virus inactivation and time-of-addition assays on cell cultures. Results: V.nilotica chloroform, methanolic and water bark extracts exerted antiviral activity against HSV-2 and HPV-16 PsV infections; among these, methanolic extract showed the best EC50s with values of 4.71 and 1.80 mu g/ ml against HSV-2 and HPV-16, respectively, and it was also active against an acyclovir-resistant HSV-2 strain with an EC50 of 6.71 mu g/ml. By contrast, no suppression of HIV infection was observed. Investigation of the mechanism of action revealed that the methanolic extract directly inactivated the infectivity of the HPV-16 particles, whereas a partial virus inactivation and interference with virus attachment (EC50 of 2.74 mu g/ml) were both found to contribute to the anti-HSV-2 activity. Conclusions: These results support the traditional use of V.nilotica applied externally for the treatment of genital lesions. Further work remains to be done in order to identify the bioactive components.

Polyanionic Amphiphilic Dendritic Polyglycerols as Broad-Spectrum Viral Inhibitors with a Virucidal Mechanism

Summary: Heparin, known for its anticoagulant properties for almost 90 years, has also been recognized as a broad-spectrum antiviral for almost 70 years. However, its anticoagulant effects limit its use as an antiviral drug. Recent research has shown that the addition of hydrophobic linkers to heparin-like compounds can create irreversible interactions with viruses while maintaining low toxicity and broad-spectrum activity. These new compounds, when tested against herpes simplex virus 2 and respiratory syncytial virus, exhibited nanomolar activity, providing a foundation for the design of new nontoxic, broad-spectrum antivirals.

Persistent chemosensory dysfunction in a young patient with mild COVID-19 with partial recovery 15 months after the onset

Summary: A 17-year-old female patient experienced long-lasting anosmia after a mild infection, with partial recovery 15 months later. Neuroimaging did not show microvascular injury in olfactory bulbs and brain, and the patient exhibited symptoms of anosmia with parosmia phenomena and gustatory impairment. Immunocytochemistry of olfactory mucosa swabs revealed high expression of ACE2 in sustentacular cells and lower expression in neuronal-shaped cells. Further longitudinal studies are needed to investigate the persistence of chemosensory impairment.

Network-based pharmacology suggested that these compounds have various modes of action against SARS-CoV-2.

Lignin: A Sustainable Antiviral Coating Material

This study developed a low-cost and environmentally sustainable antiviral surface coating using spin-coated lignin, which effectively inactivated herpes simplex virus type 2 (>99% after 30 min). The antiviral mechanism is attributed to reactive oxygen species generated upon oxidation of lignin phenols.

Microwave-assisted extraction (MAE) showed a 70% higher yield in a shorter time compared to conventional methods, along with increased flavonoid and phenolic content. This extraction technique is an ideal tool for preserving thermolabile plant constituents and is more environmentally friendly.

Summary: This study investigated the effects of Fufangmuniziqi formula (FFMN) on Alzheimer's disease (AD) and the role of alkaloids, flavonoids, and saponins in the process. The results showed that FFMN improved cognitive and memory impairment in the AD mouse model. Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

The results showed that M. longifolia extract is rich in phenolic compounds and has no observed toxicity up to a certain dosage limit.

A study integrated metabolomics and network pharmacology to investigate the effects of Shicao in alleviating acute liver injury

Shicao showed hepatoprotective effects against acute liver injury in rats.
