Summary: The metabolism and disposition of nicotine in the brain play a crucial role in determining its exposure. A new method was developed to analyze the dynamic profiles of nicotine metabolites and neurotransmitters. Inhibiting nicotine metabolism can enhance the residence of nicotine in the brain and its local neurotransmitter effects.

Summary: In this study, a novel UHPLC-MS/MS method was established and validated for the quantification of compound 1 in plasma. The pharmacokinetic parameters and brain tissue distribution of compound 1 in rats were studied using this method. The results indicated that compound 1 exhibited linear pharmacokinetic characteristics and was able to cross the blood-brain barrier. This study provides important support for the application of compound 1 in ischemic brain injury diseases.

Summary: This study characterized the pharmacokinetics, tissue distribution, and protein binding rates of curcumol in rats. The results showed that curcumol was rapidly absorbed and eliminated in rats. High accumulation of curcumol was found in the small intestine, colon, liver, and kidney. The protein binding rate of curcumol in rat plasma was high.

Summary: The present study developed a fast and reliable method to analyze endocannabinoids and their conjugated congeners in brain tissue. The method involved homogenizing the samples and using a micro solid phase extraction procedure for clean-up. UHPLC-MS/MS was employed for the analysis, and polarity switching was applied during the run to detect conjugated forms. The method achieved low limits of quantification and demonstrated good extraction recoveries.

Summary: Maize and grass silages are important for ruminant livestock, but the presence of fungi and mycotoxins can impact the quality of silage and subsequently, milk. This study in Portugal found common fungal species and analyzed their mycotoxin profiles. Fusarium mycotoxins were detected at low levels, while Penicillium and Aspergillus toxins were less prevalent in the silages studied.

Summary: The detection and understanding of emerging contaminants (ECs) pose new challenges to water pollution control. Triclosan (TCS), an emerging contaminant widely found in the environment, is a commonly used antibacterial agent. Microdialysis (MD), a sampling technique, combined with UHPLC-QTOF/MS analysis, was used to identify endogenous metabolites as biomarkers of TCS exposure in living crucian carp. The quantification of these biomarkers using UHPLC-MS/MS provided continuous monitoring of the effect of TCS on endogenous metabolites, contributing to a better understanding of its toxicological effect.

Summary: Cannabidiol (CBD) has anti-inflammatory, anxiolytic, antiseizure, and neuroprotective properties without addictive or psychotropic side effects. A VAMS method combined with ultra-high-performance liquid chromatography coupled with mass spectrometry was used to quantify CBD, THC, and their respective metabolites. The highest concentration was found in CBD-7-oic acid (7-COOH-CBD), followed by 7-hydroxy-cannabidiol (7-OH-CBD) and CBD. This analysis method is useful for studying epilepsy patients treated with CBD.

Summary: The analysis utilized LC-MS/MS technology to profile the normal human proteome from 117 samples across 46 normal tissues and organs, quantifying 10,438 unique proteins. By combining data from biological fluid proteomics of healthy individuals, tandem mass spectra corresponding to 13,028 unique human protein-coding genes were generated, serving as an important complementary resource for future biomarker discovery.

Summary: Modeling censored survival data involves estimating the conditional hazard function through parametric or semiparametric models, with the latter allowing for a weaker assumption of hazard proportionality. Kernel estimation can verify assumptions and capture changes in the hazard function over time and covariates.

Summary: The advent of high-resolution imaging has made data on surface shape widespread, leading to the need for a functional approach in shape analysis. Three innovative forms of analysis are introduced, including surface integration, focusing on sub-spaces with interesting behavior, and comparing individual shapes with a relevant control set. These ideas are particularly useful in medical contexts for assessing individual patients.
