PCL/PVA nanofibrous scaffold improve insulin-producing cells generation from human induced pluripotent stem cells

Pancreatic differentiation of stem cells will aid treatment of patients with type I diabetes mellitus (T1DM). Synthetic biopolymers utilization provided extracellular matrix (ECM) and desired attributes in vitro to enhance conditions for stem cells proliferation, attachment and differentiation. A mixture of polycaprolactone and polyvinyl alcohol (PCL/PVA)-based scaffold, could establish an in vitro three-dimensional (3D) culture model. The objective of this study was investigation of the human induced pluripotent stem cells (hiPSCs) differentiation capacity to insulin-producing cells (IPCs) in 3D culture were compared with conventional culture (2D) groups evaluated at the mRNA and protein levels by quantitative PCR and immunofluorescence assay, respectively. The functionality of differentiated IPCs was assessed by C-peptide and insulin release in response to glucose stimulation test. Real-Time PCR results showed that iPSCs-IPCs expressed pancreas-specific transcription factors (Insulin, Pdxl, Glucagon, Glut2 and Ngn3). The expressions of these transcription factors in PCL/PVA scaffold were higher than 2D groups. In addition to IPCs specific markers were detected by immunochemistry. These cells in both groups secreted insulin and C-peptide in a glucose challenge test by ELISA showing in vitro maturation. The results of current study demonstrated that enhanced differentiation of IPCs from hiPSCs could be result of PCL/PVA nanofibrous scaffolds. In conclusion, this research could provide a new approach to beta like cells replacement therapies and pancreatic tissue engineering for T1DM in the future.

The effect of source animal age, decellularization protocol, and sterilization method on bovine acellular dermal matrix as a scaffold for wound healing and skin regeneration

Summary: This study investigated the effects of animal age, decellularization protocols, and sterilization methods on xenogeneic acellular dermal matrices (ADMs) for skin regeneration. The results show that fetal source and EO-sterilized samples performed better in terms of morphology, mechanical properties, and wound treatment. There was no added value in using enzymatic treatment during the decellularization process. The findings suggest that the selection of animal age, decellularization, and sterilization methods should be based on clinical requirements.

Summary: This study evaluated the differentiation of iPSCs into IPCs on a 3D scaffold and compared it with a 2D cultured group. Expression analysis of circulating miR-22, miR-122, miR-217 and miR-367 as promising biomarkers of acute lymphoblastic leukemia

Summary: This study aimed to determine the expression levels of miRNAs in patients with acute lymphoblastic leukemia (ALL) and evaluate their value as biomarkers or therapeutic targets. The results showed that miR-217 was significantly decreased, while miR-22, miR-122, and miR-367 were increased. ROC analysis demonstrated that miR-217 and miR-367 could differentiate ALL patients from healthy individuals.

Loss of p53 activates thyroid hormone via type 2 deiodinase and enhances DNA damage

Summary: In this study, the authors investigate the role of thyroid hormones in tumor progression and identify the thyroid hormone activating enzyme D2 as a p53 target gene. They find that p53, as a cell stress sensor and tumor suppressor, suppresses D2 expression, leading to decreased availability of intracellular thyroid hormones. Conversely, loss of p53 enhances D2/TH signaling, promoting the fitness of tumor cells through modulation of genes involved in DNA damage and repair and redox signaling. In vivo deletion of D2 significantly reduces cancer progression, suggesting that targeting thyroid hormones could be a general approach to reduce invasiveness in p53-mutated neoplasms.

The Effect of Fetal Bovine Acellular Dermal Matrix Seeded with Wharton's Jelly Mesenchymal Stem Cells for Healing Full-Thickness Skin Wounds

Summary: Combining fetal bovine acellular dermal matrix (FADM) with human Wharton's jelly mesenchymal stem cells (hWJ-MSCs) can promote the healing of full-thickness skin wounds. The FADM-WJMSC group showed a higher wound closure rate and fewer inflammatory cells compared to the control and FADM groups. These findings suggest the potential application value of combining FADM with hWJ-MSCs in the treatment of skin wounds.

The androgen-thyroid hormone crosstalk in prostate cancer and the clinical implications

Summary: There is increasing evidence that thyroid hormones and androgen receptors work together to regulate gonadal differentiation and reproductive function. These hormones have interactions at the promoter region and increase expression of androgen receptors. The crosstalk between thyroid hormones and androgen receptors plays a role in the regulation of enzymes involved in androgen biosynthesis, indicating a mutual interaction. This interaction supports the relationship between hyperthyroidism and the high incidence of prostate cancer (PCa).

Summary: The anticancer activity of AMP-WF3 peptide from Poecilia Mexicana fish was evaluated against leukemic cell line Jurkat, and its cytotoxicity compared with normal cells. AMP-WF3 exhibited higher cytotoxicity against Jurkat cells with minimal effects on normal cells. The apoptotic activity of AMP-WF3 on Jurkat cells was significantly higher than that on HDF and PBMCs. The expression of P21, P53, and BCL-2 genes showed different changes in Jurkat cells, PBMCs, and HDF cells.

Comparative alternative polyadenylation profiles in differentiated adipocytes of subcutaneous and intramuscular fat tissue in cattle

Genome-wide regulation of Pol II, FACT, and Spt6 occupancies by RSC in Saccharomyces cerevisiae

Summary: RSC is an essential ATP-dependent chromatin remodeling complex in Saccharomyces cerevisiae. It regulates nucleosome-depleted regions (NDRs) by sliding flanking nucleosomes away from NDRs. Depletion of RSC leads to nucleosome encroachment in NDRs and transcription initiation defects. The study compared the effects of catalytic-dead Sth1 and rapid depletion of Sth1 on transcription. Rapid depletion of Sth1 reduces recruitment of TBP and Pol II, while the catalytic-dead mutant exhibits a severe reduction in TBP binding but accumulates Pol II in coding regions. The results suggest a role for RSC in transcription elongation and termination processes.

miR-124-3p regulates the involvement of Ptpn1 in testicular development and spermatogenesis in mouse

Age-different BMSCs-derived exosomes accelerate tendon-bone interface healing in rotator cuff tears model
