Classic SRY-box protein SOX7 functions as a tumor suppressor regulating WNT signaling and is methylated in renal cell carcinoma

SOX7 (SRY-related high mobility group box 7), a high mobility group protein, is reported to be down-regulated in several cancer types, which indicates an important role in tumorigenesis; however, its biologic role in renal cell carcinoma (RCC) pathogenesis remains unknown. We studied the alterations and functions of SOX7 in RCC. We detected its broad expression in multiple human normal tissues, including kidney, but frequent down-regulation in RCC cell lines and primary tumors. Promoter CpG methylation seems to directly mediate SOX7 silencing in RCC cells, which could be reversed by demethylation treatment. SOX7 methylation was detected in primary RCC tumors, but rarely in normal kidney tissues. Restoration of SOX7 in silenced 786-O and A498 RCC cell lines inhibited their cell growth by inducing G(0)/G(1) arrest, whereas SOX7 knockdown promoted RCC cell proliferation. We also found that SOX7 silencing resulted in the activation of WNT signaling and the induction of epithelial to mesenchymal transition. Summary: This study evaluated the potential tumor suppressor gene ZBTB28 as a marker for cervical cancer assessment. The research found that ZBTB28 can induce autophagy-related apoptosis in cervical cancer cells, inhibiting tumor growth. Furthermore, ZBTB28 promoter methylation analysis may offer a new objective strategy for cervical cancer screening.

Summary: This study reveals that HAND1 is frequently downregulated in gastric cancer due to promoter methylation, and its low expression is correlated with tumor progression and poor prognosis. High expression of HAND1 is associated with higher 5-year overall survival rates in gastric cancer patients. HAND1 inhibits gastric cancer cell growth and migration through induction of endoplasmic reticulum (ER) stress and apoptosis. The methylation status of HAND1 promoter could be a potential biomarker for gastric cancer.

Summary: Peptides/small proteins encoded by noncanonical open reading frames (ORF) of previously claimed non-coding RNAs have important biological functions but are largely uncharacterized. In this study, we identified and validated a small protein SP0495 as a novel tumor suppressor, frequently inactivated by promoter methylation in multiple tumors. SP0495 functions as a phosphoinositide-binding protein, regulating AKT signaling activation and autophagy. It inhibits tumor cell growth and induces apoptosis, cell cycle arrest, senescence, and autophagy through multiple mechanisms, including the modulation of phosphoinositides turnover and autophagic/proteasomal degradation of autophagy regulators. SP0495 has the potential to be a biomarker for cancer diagnosis and prognosis.

Selenite as a dual apoptotic and ferroptotic agent synergizes with EGFR and KRAS inhibitors with epigenetic interference

Summary: Selenite is found to induce both apoptosis and ferroptosis in lung cancer cells, especially those with EGFR and KRAS mutations. It activates the p38-ATF4-DDIT3 pathway for ferroptosis induction and influences DNA methylation machinery. Combining selenite with osimertinib or adagrasib enhances the therapeutic effect.
