Recently in Cell, Mohamed et al. (2018) report a cell-cycle regulator gene cocktail identified from young cardiomyocytes that enables mouse, rat, and human cardiomyocyte proliferation and promotes heart regeneration after infarction, defying the non-dividing nature of adult mammalian cardiomyocytes and implying a new way to treat or prevent heart failure.

Summary: The study introduces a novel in vitro platform with patterned extracellular matrix that promotes the alignment of hPSC-CMs, enabling the formation of a functional cardiac syncytium. Advances in Biomimetic Nanoparticles for Targeted Cancer Therapy and Diagnosis

Summary: The development of biomimetic nanoparticles as a novel drug delivery platform to enhance drug biocompatibility and specificity, particularly in the tumour microenvironment, has been discussed. By integrating different types of biomaterials onto the surface of nanoparticles to mimic the natural biological features and functions of cells, these nanoparticles can evade clearance by the immune system and reduce potential toxic side effects. Despite rapid progress in this field, there is still a need to advance biomimetic-based nanomedicine for clinical use, especially in the context of malignant tumors.

The presence of FN in the ECM is essential for effective cardiac differentiation, while other ECM proteins like laminin-111 and laminin-521 inhibit this process. Inhibition of certain integrin receptors and integrin-linked kinase can also impair cardiac differentiation.

Opportunities and Challenges of Human IPSC Technology in Kidney Disease Research

This review examines the development of iPSC reprogramming and characterization technologies, provides an updated list of stem cell biobanks, and highlights important iPSC-based research on diseases such as autosomal dominant kidney disease and SARS-CoV-2 kidney involvement. The review also discusses challenges and future perspectives.

Neutral sphingomyelinase regulates mechanotransduction in human engineered cardiac tissues and mouse hearts

Summary: Cardiovascular disease, the leading cause of death in the USA, is aggravated by elevated mechanical stress from hypertension. This study investigates the loss of caveolae, plasma membrane structures that buffer mechanical stress, in pathological conditions associated with chronically stretched myocardium. The findings identify neutral sphingomyelinases (nSMase) as mechanoregulated mediators of caveolae loss, ceramide production, and blunted beta-adrenergic response in a human cardiac model.
