Identifying members of the intestinal microbiota that play causal roles in shaping human disease susceptibility has been an ongoing challenge. In their recent paper in Nature, Surana and Kasper (2017) present a new strategy that may help triangulate'' causal and therapeutically useful bacteria from the human gut.

Summary: This study reveals that within-host evolution of gut commensal bacteria plays a critical role in bacterial translocation and initiation of inflammation. Gut microbiota strains can adapt and evolve, affecting their propensity to elicit inflammatory diseases. The changes in bacterial behavior are associated with genetic mutations, altered microbial gene expression programs, and remodelled cell wall structures.

Summary: Research on the microbiota-gut-brain axis has shown the crucial interactions between gut microbiota, the immune system, and the brain in various diseases and disorders. Modern studies focus on neuro-immune-microbiota interactions and understanding the mechanisms underlying the diverse impacts of the microbiome on human health. This research may lead to new approaches for treating diseases influenced by the complex connections between microbiota and the brain in the future.

Summary: Through the culturomics study on bacterial species associated with inflammatory bowel disease, two isolates tentatively identified as members of the family Erysipelotrichaceae were found to be closely related to Allobaculum stercoricanis. These isolates, strain 128T and 539T, showed strict anaerobic growth in long chains at temperatures between 37 and 42 degrees Celsius.

Highly multiplexed bioactivity screening reveals human and microbiota metabolome-GPCRome interactions

Summary: The study developed a highly multiplexed screening technology called PRESTO-Salsa that can evaluate nearly all conventional GPCRs in a single well of a 96-well plate. Using this technology, researchers screened 1,041 human-associated metabolites and discovered previously unreported GPCR agonists. They also generated an atlas of microbiome-GPCR interactions across multiple body sites, revealing conserved patterns of GPCR engagement and activation. These findings contribute to a better understanding of metabolite-GPCR interactions.

Identification of Allobaculum mucolyticum as a novel human intestinal mucin degrader

Summary: The human gut microbiota plays a key role in intestinal health and disease, with some bacteria linked to human diseases exploiting the inner mucus layer as an important niche. A newly identified IBD-associated species, Allobaculum mucolyticum, was found to carry up to 60 genes encoding putative Carbohydrate Active Enzymes (CAZymes) that may contribute to mucosal colonization. Mass spectrometry revealed the presence of 49 CAZymes, with 26 significantly enriched in its secretome, suggesting that A. mucolyticum's CAZyme secretion may facilitate bacterial colonization and degradation of the mucus layer.
