Compound 20 reduced thymocyte numbers and IL-17A containing gamma delta-T cells, and progressed to phase 1 clinical studies based on favorable safety profile.

Summary: Retinoic Acid Receptor-Related Orphan Receptor gamma t (ROR γ t) has been proposed as a potential target for the treatment of inflammatory and autoimmune diseases by regulating the production of interleukin-17 (IL-17) by T helper 17 (Th17) cells. In this study, a series of triazine-based derivatives were developed as novel ROR γ t inverse agonists. Compound 14g exhibited potent ROR γ t inverse agonistic activity and showed promising therapeutic potential in treating autoimmune diseases.

Summary: Molecular docking is a crucial method in virtual screening for identifying small-molecule ligands. However, current docking algorithms struggle to distinguish active ligands from inactive ones. This study introduces a novel docking and shape-focused pharmacophore virtual screening protocol, using retinoic acid receptor-related orphan receptor gamma t (RORγt) as a case study. The results demonstrate that this protocol achieves an effective hit rate of approximately 29%.

T(H)17 cells play a significant role in these diseases, and nuclear receptors REV-ERB alpha and beta have been identified as potential therapeutic targets for suppressing T(H)17 cell development and pathogenicity. A novel REV-ERB alpha-selective scaffold was studied in a mouse model, demonstrating its efficacy in targeting the receptor.

Summary: ROR?t, a transcription factor, is a potential drug target for autoimmune diseases due to its involvement in the differentiation of human Th17 cells. Allosteric regulation of ROR?t by small molecules has gained interest for its unique mode of action, bioactivity, and improved selectivity. This review discusses the discovery and identification of the allosteric pocket of ROR?t and provides examples of small molecules that allosterically inhibit ROR?t, including their SAR, biological activity, PK properties, and ligand binding mode. The potential role of ROR?t allosteric inverse agonists as small molecule therapeutics for autoimmune diseases is also explored.
