Summary: This study found that early tumor shrinkage (ETS) and depth of response (DoR) were significantly associated with survival outcomes in patients with MSI-H/dMMR mCRC treated with immune checkpoint inhibitors (ICIs), suggesting that they may serve as important prognostic factors for designing treatment strategies. Prospective validation of these findings is recommended.

Summary: This study investigated the relationship between gut microbiome and clinical response to anti-PD-1 immunotherapy in patients with advanced hepatobiliary cancers. Taxonomic signatures enriched in responders were effective biomarkers to predict the clinical response and survival benefit of immunotherapy. Functional annotation indicated that taxa associated with different metabolic pathways may modulate the clinical response to immunotherapy in hepatobiliary cancers.

Summary: This case series describes five patients with ICI-treated MSI mCRC with disease progression limited to the adrenal glands. The treatment methods included adrenalectomy and stereotactic body radiation therapy, suggesting that the adrenal glands may be the sanctuary sites for ICI-treated MSI mCRC.

Summary: This study used digital spatial profiling to analyze the tumor microenvironment in renal cell carcinoma (RCC) and found lower levels of immune activation markers and changes in macrophages in RCC metastases, including brain metastases. These findings have important implications for future biomarker and treatment studies.

Summary: The oncologic benefit of postchemotherapy residual tumor resection (RTR) in patients with germ cell tumors and elevated serum tumor markers (STMs) remains unclear. Analysis of the data showed that a preoperative serum level of AFP >= 30 mu g/l and viable cancer in the RTR specimens were significant predictors of relapse after first line chemotherapy. In addition, the prognosis of patients with elevated markers and surgery after second or later line therapy is poor regardless of the tumor marker levels.

Summary: Tertiary lymphoid structures (TLSs) in colorectal cancer liver metastases (CRCLM) are found to be associated with prognosis. The study analyzed data from 603 CRCLM patients and found that higher intratumor TLSs score is correlated with better prognosis, while higher peritumor TLSs score is correlated with poorer survival. Additionally, the study also found a correlation between immune cell composition within TLSs and TLSs score.

Summary: In this study of 331 patients with metastatic colorectal cancer treated with immune checkpoint inhibitors, it was found that certain immune-related adverse events (irAEs) could affect patient survival, with some having a protective effect and others having a harmful effect. The study proposed a simple model to evaluate the impact of irAEs on treatment outcomes.

Summary: CMTM6 is upregulated in colorectal cancer and is associated with lower pT stage, increased CD4(+)/CD8(+) tumor-infiltrating lymphocytes, and a favorable prognosis. PD-L1 is expressed at a low level in tumor tissues, and PD-L1 positivity in tumor stroma is associated with increased CD4(+) TIL density and a favorable prognosis. The coexpression status of CMTM6 and PD-L1 can stratify patients with CRC into different risk groups and may be especially useful for patients undergoing adjuvant chemotherapy.

Summary: This study provides evidence that left-sided colorectal cancer has a significantly better prognosis compared to right-sided colorectal cancer, in all stages and ages combined. The survival of left-sided colorectal cancer is also longer than right-sided colorectal cancer in stage IV, regardless of MMR, RAS, and BRAF status.

Summary: This study confirms that left-sided colorectal cancer has a better prognosis compared to right-sided colorectal cancer in the Belgian population. The presence of biomarkers such as MMR, BRAF, and RAS status does not significantly affect the prognostic effect of primary tumor location.

Summary: Exploring plasma proteins on a large scale is challenging, but a multi-nanoparticle workflow proves superior to conventional methods in terms of depth and precision. The physicochemical properties and surface functionalization of nanoparticles are found to affect their selectivity for specific proteins, presenting new possibilities for designing multi-nanoparticle panels in complex biological sample analysis.
