Summary: The CDK4/6 inhibitor palbociclib, alone or in combination with ruxolitinib, shows significant efficacy in reducing leukocytosis, splenomegaly, and bone marrow fibrosis in murine models of myelofibrosis, especially in inhibiting Jak2V617F mutant hematopoietic progenitors. This drug combination could have therapeutic potential for the treatment of myelofibrosis and warrants further clinical investigation.

Summary: FAM13A gene plays a crucial role in COPD, interacting with TGF-beta 1 signaling and affecting the expression of airway epithelial cells. Overexpression of FAM13A can partially protect airway tissues, potentially serving as a therapeutic approach to reduce the extent of airway remodeling.

Summary: Idiopathic pulmonary fibrosis is a chronic lung disease characterized by a decline in lung function and poor prognosis. Type I collagen, a major component of the extracellular matrix, is a potential target for fibrosis treatment. HSP47, a collagen-binding protein, is increased in pulmonary fibrosis and has implications in the development of therapeutic strategies.

Summary: Dysphagia is a common comorbidity of COPD, and it can be detected early as a manifestation of breathing-swallowing discoordination. Low-pressure CPAP and IFC-TESS have shown potential in counteracting swallowing disorders and reducing exacerbations in COPD patients. Further studies are needed to evaluate the impact of these interventions on COPD exacerbations.

Summary: The circadian clock plays a crucial role in integrating external environmental changes and internal physiology, providing temporal precision and robust adaptation to the environment. Disruption of circadian rhythms can lead to adverse health consequences, but there is still an underappreciation of how to take advantage of biological timing for health benefits. Promising progress in research and clinical applications should drive efforts towards a new era of circadian medicine.

Summary: SIRT1, as a member of the class III family of nicotinamide adenine dinucleotide (NAD(+))-dependent histone deacetylases, plays a crucial role in the progression of various diseases, including autoimmune diseases. Modulating the functions of SIRT1 may serve as a potential therapeutic strategy for these diseases.

Summary: CD47 is a protein that is ubiquitously expressed on the surface of cells and plays a critical role in self-recognition. It modulates cellular phagocytosis, determines the lifespan of erythrocytes, regulates immune cell activation, and manipulates synaptic pruning during neuronal development. Recently, CD47 has been identified as a novel target for cancer immunotherapy. This review discusses the diverse functions of CD47, its role in immune system homeostasis, and its potential therapeutic roles against cancer.

Summary: ENPP1 is frequently overexpressed in local relapses and tumor metastases, promoting an immunosuppressive tumor micro-environment and impairing immune response. Blocking ENPP1 could enhance immune remodeling and activate the STING pathway. Combination therapy with ENPP1 inhibitors and other modalities shows potential in improving antitumor-immune responses and clinical outcomes. This review summarizes the current state of the art and provides new perspectives for novel treatment strategies.

Summary: Exaggerated inflammatory response is associated with various disorders, and ATP and Panx1 channels play important roles in regulating the inflammatory response. Inhibiting Panx1 channels can alleviate inflammation and improve inflammatory diseases in multiple organs. However, most current drugs lack specificity and stability, and new drugs targeting Panx1 channels need to be developed.
