Summary: This study found that left ventricular hypertrophy increases the density of late sodium current, leading to early afterdepolarizations in the left ventricular endocardium and resulting in macroscopic T wave alternans and mechanical alternans. INa-L blockers can eliminate macroscopic T wave alternans, mechanical alternans, early afterdepolarization-induced R-T extrasystoles, and Torsades de Pointes.

Offspring of these women have a 50% risk of inheriting the disease, necessitating complex delivery room preparation in pregnancies with inherited arrhythmia syndromes. Fetal magnetocardiography now allows for the detection of various arrhythmias in susceptible fetuses, requiring specialists to have optimal knowledge and equipment for the care of these specialized pregnancies.

Summary: The establishment of human iPSC-CMs has provided a human-based platform for predicting cardiotoxicity. A novel in vitro multiple cardiac ion channel screening-based method using human iPSC-CMs was developed to predict drug-induced arrhythmogenic risk. By studying the effects of different drugs on the cardiac action potential and ion channels in human iPSC-CMs, it was found that sotalol, chlorpromazine, and mexiletine have different effects on the AP waveform and ion channels, which are associated with the risk of ventricular tachycardia TdP.
