The subcellular compartmentalization of TGFβ-RII and the dynamics of endosomal formation during the signaling events: An in vivo study on rat mesothelial cells

We previously showed that intraperitoneal administration of Freund's adjuvant treatment resulted in acute peritonitis and TGF-beta was found to be one of the main organizers of the subsequent EMT in mesothelial cells. In the present study, we investigated whether TGF-beta signaling molecules are present in mesothelial cells and how their compartmentalization pattern changes with the dynamics of inflammatory events in vivo. In addition, we tried to evaluate the turnover of endosomal compartments concomitant with the internalization of signaling molecules and examine whether caveola-mediated internalization might play a role in the termination of TGF-beta signaling. Using immunocytochemical approach, we could detect T beta RII in EEA1 positive compartments and as the inflammation progressed, at D3, the receptor appeared in caveolin-1 positive intracellular structures as well. The latter event was accompanied by the appearance of negative regulatory protein, Smad7 in caveolae. We also found EEA1 and caveolin-1 double positive vesicular structures that were corresponded to forming MVBs affirmed by our immuno-electron microscopical results. Fine structural, morphometric and immunoblot analysis proved that Cd63 positive multivesicular body (MVB) formation was significantly increased by D3 and the IP results confirmed that T beta RII as well as caveolin-1 were strongly associated with these endosomal compartments at this time. In contrast, by the termination of inflammation, by D5, caveolin-1 was found to be associated with late endosomal marker, Rab7 and entirely degraded from the system. Despite the limitations of an in vivo system, our results provide both morphological and biochemical data about the endosomal compartments involved in the internalization of T beta RII upon inflammatory stimuli. Furthermore, our study implies the possible role of caveola-mediated endocytosis in the attenuation of TGF-beta signaling and highlight the significance of endosomal compartments via which caveolae might meet the classical endocytic pathway under in vivo inflammatory conditions. Application of Multilayer Evidence for Annotation of C-Terminal BRCA2 Variants

Summary: The potential pathogenic role of germline BRCA2 c.9976A>T and c.10095delinsGAATTATATCT variants in hereditary breast and ovarian cancer (HBOC) patients was evaluated. Both variants showed no significant association with clinicopathological parameters or elevated risk for HBOC tumors. Families carrying c.9976A>T were more prone to lung cancer, while an increased prevalence of pancreatic cancer was found in families with c.9976A>T and other pathogenic BRCA1 variants. Further research in larger sample cohorts is needed for validation.

Analytical Performance of NGS-Based Molecular Genetic Tests Used in the Diagnostic Workflow of Pheochromocytoma/Paraganglioma

Summary: The study demonstrates the evolution and effectiveness of Next Generation Sequencing (NGS) techniques, particularly whole exome sequencing and targeted panel sequencing, in diagnosing hereditary cancer syndromes. NGS-based methods are efficient and cost-effective tools for molecular genetic diagnosis, especially for assessing mutations in pheochromocytoma/paraganglioma (PPGL) associated genes. Different library preparations, sequencing platforms, and bioinformatical settings significantly impact the diagnostic accuracy, with an in-house developed targeted panel sequencing showing potential for identifying genetic susceptibility of PPGL.

Wnt-Signaling Regulated by Glucocorticoid-Induced miRNAs

Summary: The study found that glucocorticoid treatment led to miRNA expression changes in different cell types. 19.5% of the influenced signaling pathways were common in all experiments, with the Wnt signaling pathway being the most affected. Transcriptome data and tissue-specific target prediction showed similar results, indicating significant alterations in the Wnt pathway in both the GC-producing adrenal gland and all investigated GC target tissue types.

Exploratory Circular RNA Profiling in Adrenocortical Tumors

This study investigated the expression profiles of circRNAs in adrenocortical tumors using next-generation sequencing and RT-qPCR validation, and found that circPHC3 is overexpressed in carcinoma and adenoma. This is the first study to investigate circRNAs in adrenocortical tumors, and further research is needed to understand their biological and diagnostic significance.

Context-Dependent Role of Glucocorticoid Receptor Alpha and Beta in Breast Cancer Cell Behaviour

Summary: This study aimed to uncover the context-dependent action of glucocorticoid receptors (GR) in breast cancer. Through multiple cohorts, it was found that GR expression is higher in ER- breast cancer cells and GR-regulated genes are mainly involved in cell migration. The study also revealed that GR action is independent of ligand availability, highlighting its ligand-independent role in breast cancer. Therefore, caution should be applied in the interpretation of immunohistochemistry results, and the importance of GR in disease progression, particularly in cell migration, is emphasized.

PALB2 Variants Extend the Mutational Profile of Hungarian Patients with Breast and Ovarian Cancer

Summary: The detection rate and characteristics of pathogenic/likely pathogenic (P/LP) variants in PALB2 gene may vary between different populations. In this study, PALB2 was analyzed in three independent cohorts and P/LP PALB2 variants were found in 1.4% of breast and/or ovarian cancer patients and in only 0.17% of patients with other cancers. The most common variant in our patients was NM_024675.4:c.509_510delGA.

Oxidative stress and signaling through EGFR and PKA pathways converge on the nuclear transport factor RanBP1
