Photoacoustic imaging is the latest promising diagnostic tool with many advantages, such as high spatial resolution, deep penetration depth, and use of non-ionizing radiation. Tumor therapy using visual guidance of photoacoustic imaging has been recently highlighted for personalized and effective tumor therapy. The goal of this study was to develop a nano-medicine tool for simultaneous photoacoustic imaging-based diagnosis and chemotherapy. Here, indocyanine green and paclitaxel doubly-loaded human serum albumin nanoparticles (IPHSA-NPs) were developed as photoacoustic imaging-based theragnostic nano-medicines. The IPHSA-NPs were on average 188.14 +/- 42.82 nm in diameter. The IPHSA-NPs were taken up by cancer cells efficiently, resulting in effective anti-cancer activity (50% less cell viability with 200 nM of paclitaxel for 6 h compared to control). The IPHSA-NPs sufficiently demonstrated photoacoustic intensity in the near infrared region at a wavelength of 800 nm. The photoacoustic intensity of IPHSA-NPs was 1.5 times stronger than hemoglobin, an endogenous photoacoustic contrast molecule. In addition, both hypodermically and intravenously administered IPHSA-NPs provided sufficient photoacoustic contrast signal intensities in in vivo over 24 h. Finally, intravenously administered IPHSA-NPs successfully accumulated in the tumor region, resulting in high photoacoustic contrast intensity. In conclusion, IPHSA-NPs are capable of photoacoustic-based cancer diagnosis and chemotherapy.

Effectiveness of synthetic aperture focusing and coherence factor weighting for intravascular ultrasound imaging

Sungwoo Kang, Junsu Lee, Jin Ho Chang

Summary: This study explores the effectiveness of using SAF and CFW methods for improving ultrasound IVUS imaging. It was found that applying CFW to the target scanline itself is beneficial for spatial resolution, while SAF is advantageous for enhancing the contrast-to-noise ratio. However, using focused IVUS transducers without these methods may be more advantageous for improving spatial and contrast resolution simultaneously due to system complexity.

Summary: In this study, tumor-derived reassembled exosome (R-Exo) was used both as a drug delivery carrier and an immunostimulatory agent for cancer therapy. By loading chlorin e6 photosensitizer into R-Exo, it retained its original size and membrane proteins for targeting tumor cells. The modified exosomes were able to generate reactive oxygen species and increase cytokine release from immune cells, showing their potential as an immunotherapeutic agent.

Elimination of Nontargeted Photoacoustic Signals for Combined Photoacoustic and Ultrasound Imaging

Haemin Kim, Hohyeon Lee, Hyuncheol Kim, Jin Ho Chang

Summary: Photoacoustic imaging has been in the spotlight for its ability to provide optical contrast images of physiological information and deep imaging depth, but its sensitivity is limited by nontargeted signals and clutter artifacts. Researchers proposed a method to eliminate nontargeted signals using a dual-modal ultrasound-photoacoustic contrast agent and demonstrated improved visualization of target areas in photoacoustic images.
