Somatic Host Cell Alterations in HPV Carcinogenesis

High-risk human papilloma virus (HPV) infections cause cancers in different organ sites, most commonly cervical and head and neck cancers. While carcinogenesis is initiated by two viral oncoproteins, E6 and E7, increasing evidence shows the importance of specific somatic events in host cells for malignant transformation. HPV-driven cancers share characteristic somatic changes, including apolipoprotein B mRNA editing catalytic polypeptide-like (APOBEC)-driven mutations and genomic instability leading to copy number variations and large chromosomal rearrangements. HPV-associated cancers have recurrent somatic mutations in phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha (PIK3CA) and phosphatase and tensin homolog (PTEN), human leukocyte antigen A and B (HLA-A and HLA-B)-A/B, and the transforming growth factor beta (TGF beta) pathway, and rarely have mutations in the tumor protein p53 (TP53) and RB transcriptional corepressor 1 (RB1) tumor suppressor genes. There are some variations by tumor site, such as NOTCH1 mutations which are primarily found in head and neck cancers. Understanding the somatic events following HPV infection and persistence can aid the development of early detection biomarkers, particularly when mutations in precancers are characterized. Somatic mutations may also influence prognosis and treatment decisions.

Summary: This study showed that early-relapse hepatocellular carcinoma (HCC) tumors exhibit innate-like CD8(+) T cells with overexpression of KLRB1 (CD161) and a low cytotoxic state, unlike the exhausted state seen in primary HCC. These cells were associated with a worse prognosis. Additionally, potential immune evasion mechanisms were identified in recurrent tumor cells, which dampen dendritic cell antigen presentation and recruit innate-like CD8(+) T cells.

scDPN for High-throughput Single-cell CNV Detection to Uncover Clonal Evolution During HCC Recurrence

Summary: The single-cell DNA library preparation method developed in this study, known as scDPN, eliminates the need for preamplification and allows for high-throughput CNV detection. It shows lower amplification bias and noise compared to the MDA method, and demonstrates high sensitivity and accuracy in evaluating cell lines and tumor tissues. Additionally, it was used to identify clonal subpopulations in hepatocellular carcinoma (HCC) samples, revealing clonal selection during tumor recurrence. Overall, this approach provides a comprehensive and scalable solution for understanding genome heterogeneity and evolution.

Summary: This study aimed to determine the effect of HLA-A*03 as a biomarker for predicting response to immunotherapy. Mutations in ABC genes can lead to various human disorders and phenotypes, and mouse models with disrupted ABC genes mimic these human diseases. New genomic studies and tumor sequencing have provided additional insights into the functions of ABC genes.

Summary: E7 gene variants in HPV16 are commonly found in HPV-infected individuals without cancer. These variants show reduced activity, leading to uncontrolled cell growth and invasion. Inhibition of E7 could be a potential approach for treating HPV-driven cancers.

Summary: This study found a positive association between exposure to diesel exhaust and bladder cancer, particularly in cases with high-grade, nonmuscle invasive TP53-mutated tumors. In muscle-invasive tumors, there was also a positive association between diesel exposure and nitro-PAH signatures.

Summary: This study identified pathogenic or likely pathogenic variants in cancer-susceptibility genes not previously associated with pediatric cancer in long-term pediatric cancer survivors. Additionally, known associations between certain genes and specific types of pediatric cancers were confirmed. Further characterization of these variants in pediatric cancer is important for optimal genetic counseling for patients and families.
