MiR-26a downregulates retinoblastoma in colorectal cancer

MicroRNAs are non-coding short RNAs that target the 3' untranslated region of messenger RNAs (mRNAs) and lead to their degradation or to translational repression. Several microRNAs have been designated as oncomirs, owing to their regulating tumor suppressor genes. Interestingly, a few of them have been found to target multiple genes whose simultaneous suppression contributes to the development of a tumoral phenotype. Here, we have showed that miR-26a is overexpressed in colorectal cancer data obtained from TCGA Research Network and in human colon cancer pathological specimens; moreover, an orthotopic in vivo model of colon cancer showed overexpression of miR-26a, while Rb1 expression inversely correlated to miR-26a in TCGA Research Network data, pathological samples, and the in vivo model. Then, by means of luciferase assay, we demonstrated that miR-26a targets the 3' untranslated region of Rb1 mRNA directly. This is, to our knowledge, the first report of miR-26a targeting Rb1 in colon cancer. The results of this study suggested that miR-26a could serve as a progression biomarker in colorectal cancer. Further validation studies are still needed to confirm our findings.

The Role of Hypoxia in Endometrial Cancer

Summary: Endometrial cancer, the most frequent neoplasia from the corpus uteri, is a leading cause of death in women worldwide. Chemotherapy's clinical effectiveness is unstable, calling for novel molecular targeted therapies that can improve the success rates of endometrial cancer treatment. During tumor growth, cancer cells adapt to decreased oxygen availability by upregulating hypoxia-inducible factors, leading to enhanced cell survival, invasiveness, proliferation, and resistance to chemotherapy. Hypoxia-inducible factor 1 alpha (HIF-1 alpha), which is aberrantly overexpressed in various solid tumors, has been considered an important therapeutic target. MicroRNAs also play a significant role in regulating the protein-encoding genes involved in the hypoxia response in endometrial cancer.

Summary: Protein-protein interactions are important for predicting the function of target proteins and the ability of drugs to affect biological systems. Experimental validation confirmed the relationships among these proteins and suggested a connection between RNA biology and metabolism.

Summary: Polyphenols are secondary metabolites from plants with natural antioxidant capacity and biological activities. Due to their mostly polar character, they have low solubility in less polar environments or hydrophobic matrices. However, by functionalizing them through chemical or enzymatic lipophilization, phenolipids can be synthesized. These amphipilic molecules possess a natural phenolic core and an attached aliphatic motif, allowing them to interact with various systems and potentially be used in processed foods, as drug carriers, antimicrobial agents, antioxidants in the cosmetic industry, and even in the treatment of degenerative diseases related to oxidative stress.

Reprogramming of the Genome-Wide DNA Methylation Landscape in Three-Dimensional Cancer Cell Cultures

Summary: During the last century, 2D cell cultures have been commonly used to study cancer biology, drug discovery, genomics, and gene expression regulation. However, this approach has limitations in accurately reproducing the tumor microenvironment and cellular processes. To overcome these limitations, 3D cell cultures have been implemented to better mimic the biological and molecular features of tumors. This review discusses the application of different types of 3D cell culture models to characterize the epigenetic changes in cancer cells, particularly DNA methylation patterns, and the impact of microenvironmental factors on these changes.

Pathogenic variant profile in DNA damage response genes correlates with metastatic breast cancer progression-free survival in a Mexican-mestizo population

Summary: This study sequenced the DNA of 40 Mexican-mestizo metastatic breast cancer patients and identified 22 variants, including 9 novel ones. The study found a significantly high proportion of variations in ARID1A, and the presence of variants in ARID1A, BRCA1, BRCA2, or FANCA genes was associated with worse survival outcomes. The findings suggest routine screening for ARID1A and BRCA1/2 variants in Mexican-mestizo breast cancer patients.

Lithium: A Promising Anticancer Agent

Summary: Lithium, an anti-cancer agent, shows potential in inducing apoptosis, autophagy, and inhibiting tumor growth. It also plays a role in tumor regulation and synergizes with standard cancer therapies. Additionally, lithium has neuroprotective effects and enhances anti-tumor activities in nano-sized form. However, its therapeutic activities depend on various factors. This review emphasizes the importance of repurposing lithium as a therapeutic agent and highlights its molecular mechanisms.

Metastatic breast tumors downregulate miR-145 regulating the hypoxia-induced vasculogenic mimicry

Summary: This study evaluated the expression of VM-associated microRNAs in tumors of metastatic breast cancer patients and explored their potential therapeutic value in disease progression. It was found that certain microRNAs regulating VM and metastasis were downregulated in primary tumors of patients with metastatic disease, and these regulatory factors had predictive value in patient survival. Furthermore, in vitro experiments demonstrated that miR-145 mimics could inhibit the development of VM in metastatic breast cancer cells.

Summary: This study evaluated the role of SOX9 in colorectal cancer stem cells (CRC-CSCs). The results showed that SOX9 promotes the stemness properties of CRC-CSCs by regulating stem cell markers, increasing sphere formation, and maintaining CSCs surface markers. Therefore, SOX9 may serve as a potential therapeutic target for CRC stem cells.

Summary: Testicular cancer is the most common tumor in males aged 15 to 35, with a high number of new cases and deaths each year. Non-coding RNAs (ncRNAs) play a crucial role in the regulation of cellular processes and pathologies, including testicular cancer. Epigenetic modifications, such as histone modifications and DNA methylation, along with microRNA (miRNA) regulation, are also associated with testicular tumor progression and treatment response. This review focuses on the latest discoveries in the epigenetic aspect of testicular cancer, particularly the involvement of proteins and ncRNAs like miRNAs and lncRNA, highlighting their relevance in pathogenesis and potential for prognostic evaluation and targeted therapies.
