Summary: Mass cytometry (CyTOF) is a new technology that enables high-resolution investigation of protein expression at the single cell level. This study provides a structured and reproducible method for staining platelets from platelet-rich plasma to study thrombocyte protein expression using CyTOF, allowing for sample storage and delayed measurement. The protocol also includes a bioinformatic pipeline for analyzing platelet expression, unlocking the potential of CyTOF analysis for studying platelet biology in health and disease.

Summary: Platelet-rich plasma (PRP) is a biologic product commonly used in medicine for tissue repair. This study developed a new method to obtain PRP enriched in both platelet and extraplatelet molecules using hydroxyethyl acrylamide (HEAA)-based hydrogels. The resulting PRP showed higher levels of extraplatelet growth factors and improved cell viability compared to standard PRP. This absorption-based method has the potential to improve tissue regeneration capacity.

Summary: This study investigated the influence of kidney function on PCSK9 levels and its role in vascular smooth muscle cell (SMC) calcification. The results showed a negative association between glomerular filtration rate and plasma PCSK9 levels. Overexpression of PCSK9 in cells led to increased calcification and release of extracellular vesicles enriched in calcium and alkaline phosphatase. The findings suggest a direct role of PCSK9 in vascular calcification associated with renal impairment.

Summary: This study evaluated the impact of four COVID-19 vaccines on platelet activation, coagulation, and inflammation. It found that vaccination did not induce platelet activation and microvesicle release. Although there were minor changes in clotting activation and endothelial function, these could potentially explain the rare venous thromboembolic complications of COVID-19 vaccination at a population level.
