Summary: Abnormal activation of the immune system is a known cause of lupus. Inhibiting the inflammasome and its downstream components has been shown to reduce disease severity in lupus mouse models. This study examines the effects of IL-1 beta loss on disease severity in a lupus-prone mouse model and identifies potential clinical relevance of TNF and IL-17 signaling pathways. Caution may be needed when inhibiting inflammasome or IL-1 beta signaling in SLE, especially in patients with renal involvement.

Summary: This study found that Hyperoside can inhibit the activation of the NLRP3 inflammasome by reducing the secretion of inflammatory factors, protecting dopaminergic neurons, and reversing motor dysfunction caused by MPTP. Hyperoside achieves this by upregulating PACAP levels.

Summary: The study reveals that albumin/R-III inhibits the transforming growth factor-beta-Smad3 signaling by sequestering retinoic acid, leading to nuclear translocation of p65 and increased IL-1beta expression. Moreover, albumin/R-III-induced phosphorylation of Smad3 inhibits HSC activation, suggesting the potential anti-fibrotic effects of R-III.

Summary: Fascioliasis, a food-borne helminthiasis, primarily affects cattle and sheep but can occasionally occur in humans. This study found that the aquatic herb water dropwort is the primary source of human infections in Korea. However, a cluster of patients who did not consume water dropwort but had used a green vegetable delivery service were diagnosed with fascioliasis. Further research is needed to confirm the transmission of human fascioliasis through green vegetable juice delivered by modern industrial systems.

Summary: This study found that the plant height of water dropwort significantly increased after treatment with different concentrations of gibberellin (GA). Anatomical structure analysis indicated that GA treatment caused cell elongation in water dropwort. Metabolome analysis showed that flavonoids were the most abundant metabolites and their biosynthesis was regulated by GA.

Summary: In this study, it was found that HYP attenuated cadmium-induced kidney injury by suppressing NLRP3 inflammasome activation. This protective effect may be achieved by reducing kidney function indexes, decreasing the release of pro-inflammatory cytokines, reducing ROS production, and suppressing NLRP3 inflammasome activation. Additionally, HYP inhibited Cd-induced MAPK/NF-kappa B pathway stimulation.

Summary: The present study aimed to demonstrate the effect and mechanism of hyperoside in Doxorubicin-induced cardiotoxicity. Hyperoside attenuated the cardiac dysfunction and cardiomyocyte apoptosis induced by Doxorubicin, mainly through inhibiting the NOXs/ROS/NLRP3 inflammasome signaling pathway, which is related to oxidative stress.

Summary: This study examined the efficacy of canakinumab in patients with type 2 diabetes and obesity who were hospitalized with COVID-19. The results showed that canakinumab did not significantly improve the primary composite outcome of the patients. However, patients treated with canakinumab required less anti-diabetes drugs and experienced a prolonged reduction in systemic inflammation.

Summary: This study investigated the effects of a ketogenic diet on anthropometric indices, lipid profile, and the inhibition of NLRP3 inflammasome in obese women. The results showed that the ketogenic diet led to significant weight loss, reduced waist circumference and BHB levels, and decreased BMI and appetite. Cholesterol, triglycerides, and HDL-C slightly increased, while LDL-C and 1L-1 beta decreased significantly. The questionnaire results indicated improvements in various physical symptoms.

Summary: The deficiency of HDAC6 does not affect the release of IL-1β mediated by NLRP3 in human immune cells, indicating that the inhibition of HDAC6 is not necessary for the function of the NLRP3 pathway. In addition, HDAC6 inhibitors have the potential to suppress inflammatory responses and inhibit the release of IL-1β by regulating Toll-like receptor (TLR) signaling pathway.

The inhibition of NLRP3 triggers on LCN2 secretion is caused by the inhibited transcription and translation of LCN2, suggesting a new linkage between inflammasome activation and LCN2 secretion in the innate immunity.
