Promotion of oral surgical wound healing using autologous mucosal cell sheets

Objectives: Severe oral mucosal and tissue defects can lead to pain, infection, and later undesirable healing of scarring and adhesion, resulting in a poor quality of life. In vitro-engineered oral mucosal equivalents for covering such defects are an alternative to avoiding the donor site morbidity of conventional skin or tissue grafts. We examined the efficacy of our newly developed three-dimensional mucosal cell sheets in an in vivo tongue wound model mimicking the surgical extirpation of tongue cancer. Materials and methods: Small oral mucosal and autologous fibrin samples were obtained from surgical patients and Sprague-Dawley rats. The fibrin was mixed with fibroblasts and seeded with keratinocytes that had been primarily cultured for in vitro cell expansion. The three-dimensional autologous cell sheets, cultured in air-lift interface inserts, were transplanted into deep wounds of the rat ventral tongue. Gross and microscopic findings of the postsurgical wounds were compared between wound control and cell sheet groups. Results: The cell sheets were flexible, expandable, and easy to transfer, and had histological characteristics similar to that of the normal oral mucosa, with high p63 positivity. They promoted oral wound healing with earlier re-epithelialization and less fibrosis than that in the wound control. The cell sheet-healed tongue had similar histology to that of a normal tongue. Conclusions: Our engineered cell sheets have potential applicability for the rapid healing of oral mucosal and soft tissue defects, without scarring, adhesion, and functional deficits. Condensed abstract: The efficacy of in vitro-engineered mucosal equivalents, using completely autologous mucosa and plasma, was examined. Transplantation of the autologous cell sheets into deep wounds of the rat ventral tongue promoted oral wound healing with earlier re-epithelialization and less fibrosis than that in controls. Ferroptosis induction via targeting metabolic alterations in head and neck cancer

Summary: Ferroptosis is a newly regulated cell death induced by iron-mediated lipid peroxidation. Altered cancer metabolism contributes to cancer proliferation, metastasis, and treatment resistance. Changes in metabolism increase the sensitivity of cancer cells to lipid peroxidation toxicity and can be targeted for cancer treatment.

Lipid metabolism alterations and ferroptosis in cancer: Paving the way for solving cancer resistance

Summary: Cancer disrupts lipid metabolism, leading to altered metabolism intermediates and contributing to abnormal growth and metastasis. The shift in lipid metabolism towards an increased presence of polyunsaturated fatty acids (PUFAs) in membrane phospholipids (PLs) also causes resistance to cancer therapy. The vulnerability of cancer cells to lipid peroxidation (LPO) and subsequent ferroptosis, a new form of regulated cell death, depends on adaptive lipidome remodeling, LPO patterns, and LPO scavenging ability. Understanding the molecular basis of altered lipid metabolism in cancer may offer new opportunities for overcoming cancer resistance.

Promotion of ferroptosis in head and neck cancer with divalent metal transporter 1 inhibition or salinomycin

Summary: Divalent metal transporter 1 (DMT1) inhibitors or salinomycin can induce lysosomal iron overload and selectively kill iron-addicted cancer stem cells in head and neck cancer (HNC) cells. This study revealed that DMT1 inhibition or salinomycin treatment promotes ferroptosis by increasing labile iron levels and lipid peroxidation. Silencing of DMT1 resulted in molecular changes related to iron starvation response. These findings suggest that targeting DMT1 or using salinomycin could be a novel strategy for killing iron-avid cancer cells in HNC.

Targeting Iron-Sulfur Clusters in Cancer: Opportunities and Challenges for Ferroptosis-Based Therapy

Summary: Iron dysregulation is a characteristic of cancer, characterized by increased expression of genes involved in iron metabolism and iron-sulfur cluster (ISC) biogenesis. Dysregulated iron homeostasis leads to elevated intracellular labile iron, which can result in the formation of excessive cytotoxic radicals and susceptibility to regulated cell death, including ferroptosis. Various strategies, such as redox operations, iron chelation, and replacement of iron with redox-inert metals, can destabilize or restrict ISC formation and function, providing potential therapeutic approaches for cancer treatment. Targeting ISCs to induce ferroptosis shows promise in cancer therapy. This review provides a comprehensive overview of iron metabolism, ferroptosis, and the role of ISC modulation in cancer cells, as well as the potential of targeting ISCs for ferroptosis induction in cancer treatment. Further research and clinical trials are needed to validate these strategies for various cancers, with the goal of developing novel and effective cancer treatments.

Nrf2 targeting in overcoming ferroptosis evasion in head and neck cancer

Summary: Ferroptosis, characterized by lipid peroxidation and iron accumulation, is inhibited by the Nrf2 pathway. In head and neck cancer cells, activating the Nrf2-antioxidant responsive element pathway leads to resistance to ferroptosis, while inhibiting this pathway reverses this resistance. Modulating the Nrf2 pathway could be a promising strategy to overcome resistance in head and neck cancer therapy.

Frey syndrome after conservative parotidectomy: Importance of closing the remnant parotid parenchyma

Transoral Laser-Assisted Infrahyoid Supraglottic Laryngectomy for Selected Patients With Supraglottic Cancer

Summary: Supraglottic laryngectomy has undergone advancements from open to transoral endoscopic approaches, particularly with the use of lasers, endoscopes, ultrasonic devices, and robotics. Transoral laser-assisted microsurgery has emerged as an effective alternative to partial laryngectomy or non-surgical therapies, providing faster functional recovery. A new technique called transoral laser-assisted infrahyoid supraglottic laryngectomy allows for tumor removal while preserving crucial structures, resulting in optimal preservation of laryngopharyngeal function and minimal complications.

Comprehensive clinical, genome and transcriptomic analysis of primary ghost cell odontogenic carcinoma

Summary: This study presents a comprehensive clinical, genomic, and transcriptomic analysis of primary ghost cell odontogenic carcinoma (GCOC). The results provide the first comprehensive molecular atlas for primary GCOC, including previously identified CTNNB1 mutation and novel alterations in MAP3K, EP300, and the 22q11.21 region. The transcriptome analysis reveals significant involvement of cytokine-cytokine receptor interaction and the PI3K-Akt signaling pathway. More GCOC cases should be compared to validate these findings for accurate clinical guidance.

Transoral robotic surgery in oropharyngeal squamous cell carcinoma: A comparative study between da Vinci Single-Port and da Vinci Xi systems

Summary: This study compares the outcomes of transoral resections using the da Vinci SP and da Vinci Xi systems for oropharyngeal squamous cell carcinoma (OPSCC). The results indicate that the da Vinci SP system has advantages in terms of reduced docking and console times, as well as improved rates of wide negative resection margins. However, there were no significant differences in complication rates or postoperative functional outcomes between the two systems.
