Summary: This study analyzed the spike protein receptor-binding domain (RBD) sequence of SARS-CoV-2 isolates from COVID-19-positive patients in Odisha, India, and found a specific mutation leading to stronger binding affinity with human ACE2. The RBDs of all Indian isolates showed affinity for ACE2 of different species, suggesting potential wide infectivity and natural reservoirs for the virus.

Quantitative proteomics of hamster lung tissues infected with SARS-CoV-2 reveal host factors having implication in the disease pathogenesis and severity

Summary: This study confirmed the infectivity of a local SARS-CoV-2 isolate in a hamster model, showing similar lung pathology to COVID-19 patients and other hamsters. The lung-associated pathological changes were prominent on the 4th day post-infection and mostly resolved by 14 dpi. Proteomic analysis identified 68 significantly altered proteins between infected groups, with highlighted pathways including complement and coagulation cascade, platelet activation, ferroptosis, and focal adhesion. Additionally, altered expression of pulmonary surfactant-associated proteins Sftpd and Sftpb was observed, suggesting their potential role in SARS-CoV-2 pathogenesis and disease progression.

PM2.5 particles were classified into four categories, with the quantification of endotoxin concentrations in ambient PM2.5. Detection of heavy metals in PM2.5 was used for excess cancer risk assessment.

Gemcitabine induces polarization of mouse peritoneal macrophages towards M1-like and confers antitumor property by inducing ROS production

Summary: In patients with pancreatic cancer, the peritoneal cavity is a common site of metastasis. Peritoneal macrophages play a significant role in peritoneal metastases of different cancers. This study investigates the effect of Gemcitabine (GEM), a chemotherapy drug, on the polarization of macrophages and its potential clinical implications in pancreatic cancer. The results show that GEM induces a M1-like polarization of macrophages, resulting in cytotoxicity towards pancreatic cancer cells and delayed disease progression in a mouse model.

Summary: The study demonstrates the successful decellularization of caprine pancreas for pancreatic xenotransplantation, resulting in a decellularized pancreas tissue (DPT) with minimal immunogenicity and preserved key components like collagen and extracellular matrix. The recellularization of DPT with mouse islets showed no toxicity, retained biological and physical properties of the native ECM, making it a safer and better alternative for transplantable organs from xenogeneic sources.

Therapeutic role of N-acetyl cysteine (NAC) for the treatment and/or management of SARS-CoV-2-induced lung damage in hamster model

Summary: Oxidative stress caused by ROS is the main mediator of SARS-CoV-2-induced pathology. NAC, with its antioxidant, anti-inflammatory, mucolytic, and antiviral properties, may have beneficial effects in COVID-19 patients. This study shows that high doses of NAC can suppress severe lung damage caused by SARS-CoV-2, but fail to restrict viral load. However, high doses of NAC with and without remdesivir significantly suppress the expression of pro-inflammatory genes in lung tissues.
