Summary: Tumorigenesis and senescence, although seemingly contradictory, share similar mechanisms. Pancreatic ductal adenocarcinoma (PDA) is a lethal disease that progresses through multiple steps. Senescence is prevalent in PDA development, but also plays a significant role in promoting tumorigenesis. This review focuses on the dual roles of senescence in PDA, the signaling effectors that regulate senescence, and molecular targets for controlling senescence in PDA treatment.

Summary: Low handgrip strength and increased arterial stiffness are associated, as shown in the multivariable linear regression analysis where a decrease of 5 kg in handgrip strength led to an increase of 0.08 m/s in PWV. This suggests a connection between low handgrip strength and arterial stiffness, potentially linking to increased cardiovascular morbidity and mortality.

Summary: The functional state of T cells is critical for effective antitumor immunity. T cell senescence driven by malignant tumor cells and regulatory T cells is a common feature in cancers, characterized by unbalanced lipid metabolism and accumulation of lipid droplets. Inhibition of group IVA phospholipase A(2) can reprogram effector T cell lipid metabolism, prevent T cell senescence, and enhance antitumor immunity and immunotherapy efficacy.

Summary: This study reveals that circWDR37 deficiency limits the proliferation, migration, and invasion of senescent NPC cells induced by cisplatin or gemcitabine. Mechanistically, circWDR37 binds and dimerizes PKR, leading to PKR autophosphorylation and activation. Activated PKR phosphorylates IKKβ, releasing RELA from IκBα and activating NF-κB, thus promoting the transcription of CCND1 and senescence-associated secretory phenotype component genes in a circWDR37-dependent manner. Low circWDR37 levels are associated with favorable survival and chemotherapy response in NPC patients treated with gemcitabine or cisplatin induction chemotherapy.
