Oxidative stress in brain underlies the major neurological disorders including Alzheimer's disease (AD) and Parkinson's disease (PD). Peripherally, Angiotensin-II is a major effector of inflammation. Identification of its capacity to access brain during hypertension, as well as location of central renin angiotensin system have led to its recognition as the major effector of oxidative stress in brain. Clinical uses of antioxidants to antagonize this oxidative stress have mostly failed. In this scenario, AT1 blockers have been investigated to prevent neurodegeneration. Although it has shown promise, clinical efficacy is limited to few drugs including telmisartan mainly due to the poor brain availability of others. In this review we aim to analyze the potential of antioxidants to reduce oxidative stress in brain. We have given critical analysis of the approaches for re-purposing of AT1 blockers against oxidative stress induced neurodegeneration.

Low-volume plasma sampling for determination of dextromethorphan and dextrorphan in rat plasma: LC-MS/MS method and its application in pharmacokinetic study

Summary: Dextromethorphan (DM) and its metabolite dextrorphan (DX) have diverse pharmacodynamics, but further validation in different preclinical models and correlation with relevant pharmacokinetics data is needed. A new method using solid-phase extraction, liquid chromatographic separation, and mass spectrometric detection was developed for low sample volumes. This method allowed for the simultaneous quantification of DM and DX in rat plasma and yielded accurate pharmacokinetic parameters.

Modeling of chitosan modified PLGA atorvastatin-curcumin conjugate (AT-CU) nanoparticles, overcoming the barriers associated with PLGA: An approach for better management of atherosclerosis

Summary: Conjugate drugs are being widely studied for their potential in improving the properties of drugs. A new conjugate derivative (AT-CU) of atorvastatin and curcumin was synthesized and evaluated in terms of its physical and therapeutic properties. Chitosan-modified PLGA nanoparticles were prepared and showed improved drug encapsulation efficiency and reduced burst release. In vivo studies further confirmed the effectiveness of the ideal formulation in treating atherosclerosis.

Novel atorvastatin-curcumin conjugate nanogel, a selective COX2 inhibitor with enhanced biopharmaceutical profile: Design, synthesis, in silico, in vitro, and in vivo investigation

Summary: This study aimed to overcome solubility issues of atorvastatin (AC) and curcumin (CUR) by synthesizing atorvastatin-curcumin conjugate (AC-CUR) and evaluating its synergistic action on inflammation. The AC-CUR conjugate was successfully synthesized and found to increase solubility by approximately 2 folds. In vivo testing revealed that the new conjugate molecule exhibited excellent biopharmaceutical properties and improved anti-inflammatory efficacy, suggesting that AC-CUR nanogel could be an efficient platform for inflammation treatment.

Metal nanoparticles, synthesized using a green approach, exhibit significant anti-diabetic and antioxidant activities in a diabetic rat model.

Summary: The present study investigates the efficacy of antidiabetic drug VLD and antioxidant VE against STZ-induced diabetic cognitive impairment. To enhance the efficacy of VLD, gold nanoparticles are conjugated with VLD for targeted brain delivery. The treatment with drug-conjugated Au NPs improves cognitive functions and shows neuroprotective action against diabetes-induced cognitive impairment.

New insights toward molecular and nanotechnological approaches to antidiabetic agents for Alzheimer's disease

Summary: Alzheimer's disease (AD) and type-II diabetes mellitus (DM) are mutually related chronic neurodegenerative disorders. Chronic DM increases the risk for AD due to insulin resistance and cognitive dysfunction. Repurposing of antidiabetic drugs and nanotechnological interventions show promise in the prevention and treatment of AD. This review explores the cellular and molecular pathophysiologies of AD and type-II DM, the use of in vivo DM models in AD studies, and the potential of antidiabetic drugs and nanodelivery systems for AD treatment.

Ameliorative and Neuroprotective Effect of Core-Shell Type Se@Au Conjugated Hesperidin Nanoparticles in Diabetes-Induced Cognitive Impairment

Summary: Diabetes mellitus leads to cognitive dysfunction due to insulin deficiency. Hesperidin, a bioflavonoid, has poor solubility and a short half-life. To overcome these limitations, hesperidin was conjugated with gold, selenium, and core-shell bimetallic nanoparticles. The synthesized nanoparticles showed neuroprotective effects in a rat model of cognitive dysfunction induced by streptozotocin. Among the nanoparticles, the bimetallic Se@Au-HSP NPs exhibited the strongest neuroprotective action. Hesperidin-conjugated bimetallic nanoparticles could be a potential alternative for neurodegenerative diseases.

Metal Nanoparticles in Alzheimer's Disease

Summary: Nanotechnology is being applied in various biomedical fields, such as lifestyle diseases and neurodegenerative diseases. Metal nanoparticles are widely used as drug delivery systems due to their enhanced physicochemical properties. This review article discusses the neuroprotective and neurotoxic effects of different metal nanoparticles on Alzheimer's disease, as well as their biocompatibility for brain targeting. It also analyzes the need for new drug delivery systems and offers guidance for researchers in brain-targeted drug delivery for Alzheimer's disease.

Azilsartan ameliorates aluminium chloride induced Alzheimer's disease like pathology

Summary: This study evaluated the efficacy of Azilsartan against aluminium chloride induced Alzheimer's disease (AD). The results showed that both Azilsartan and Telmisartan significantly improved cognitive dysfunction, enhanced antioxidant status, and reduced A beta production. However, the protective effect of Azilsartan was found to be slightly weaker than Telmisartan.
