Assessment of Heparanase-Mediated Angiogenesis Using Microvascular Endothelial Cells: Identification of λ-Carrageenan Derivative as a Potent Anti Angiogenic Agent

Heparanase is overexpressed by tumor cells and degrades the extracellular matrix proteoglycans through cleavage of heparan sulfates (HS), allowing pro-angiogenic factor release and thus playing a key role in tumor angiogenesis and metastasis. Here we propose new HS analogs as potent heparanase inhibitors: Heparin as a positive control, Dextran Sulfate, lambda-Carrageenan, and modified forms of them obtained by depolymerization associated to glycol splitting (RD-GS). After heparanase activity assessment, 11 kDa RD-GS-lambda-Carrageenan emerged as the most effective heparanase inhibitor with an IC50 of 7.32 ng/mL compared to 10.7 ng/mL for the 16 kDa unfractionated heparin. The fractionated polysaccharides were then tested in a heparanase-rich medium-based in vitro model, mimicking tumor microenvironment, to determine their effect on microvascular endothelial cells (HSkMEC) angiogenesis. As a preliminary study, we identified that under hypoxic and nutrient poor conditions, MCF-7 cancer cells released much more mature heparanase in their supernatant than in normal conditions. Then a MatrigelTM assay using HSkMEC cultured under hypoxic conditions in the presence (or not) of this heparanase-rich supernatant was realized. Adding heparanase-rich media strongly enhanced angiogenic network formation with a production of twice more pseudo-vessels than with the control. When sulfated polysaccharides were tested in this angiogenesis assay, RD-GS-lambda-Carrageenan was identified as a promising anti-angiogenic agent.

Delayed alveolar clearance of nanoparticles through control of coating composition and interaction with lung surfactant protein A

Summary: The coating composition of nanomedicines plays an important role in determining the fate and clearance of the medicines in the body. In this study, two micellar superparamagnetic iron oxide nanoparticles (SPION) with different coatings were compared to understand their impact in pulmonary administration. The results showed that phosphatidylcholine (PC) coating could prolong the retention time of the nanoparticles in the lungs, making it suitable for pulmonary drug delivery. On the other hand, bovine serum albumin (BSA) coated nanoparticles were cleared more rapidly.

Summary: The study revealed that microbial mats from exploited and abandoned salterns exhibit different bacterial and meiofaunal diversity, with higher salinity in exploited sites leading to lower bacterial diversity but higher eukaryotic diversity. The presence of certain bacterial and eukaryotic groups in the mats may be attributed to human actions such as salt recovery practices in exploited salterns.

Summary: Manganese ferrite nanoparticles have unique properties in bioimaging and catalytic therapies. They have been used as contrast agents in magnetic resonance imaging (MRI) and as catalase-mimicking nanozymes for hypoxia alleviation. A cost-efficient synthetic microwave method has been developed to manufacture ultrasmall manganese ferrite nanoparticles with enhanced bioimaging and catalytic properties. These nanoparticles can be used as multimodal contrast agents in MRI and positron emission tomography (PET), and also have improved catalytic efficiency.

Summary: EBV infection is widespread and can lead to lymphomas. Both LCLs and DLBCLs cells overexpressing PD-L1 can inhibit T-cell response. Fucoidan, extracted from brown seaweed, shows potential for reducing PD-L1 expression and improving treatment of lymphoma cells. This finding has implications for immunotherapy.

Summary: Oligosaccharides derived from ?-carrageenan (?-COs) have shown promise in regulating heparanase activity and have potential therapeutic applications in cancer treatment. Commercial ?-carrageenan is a mixture of different CAR families and its composition does not reflect the real composition, limiting its clinical use. By comparing six commercial ?-CARs and applying H2O2-assisted depolymerization, ?-CO formulations with similar molar masses and sulfation degrees to those suitable for antitumor properties were obtained. However, the anti-HPSE activity of these new ?-COs was affected by other features, such as differences in the initial mixture composition and degradation induced by H2O2-based hydrolysis.

Heparin length in the coating of extremely small iron oxide nanoparticles regulates in vivo theranostic applications

Summary: This study successfully prepared HEP-ESIONP probes with excellent MRI contrast using a one-pot microwave method with heparin oligosaccharides (HO) of different lengths as coatings, demonstrating anticoagulant properties and/or specific antitumor activity based on the length of HO used. In vivo distribution study showed significant changes in probe behaviors and renal clearance with shortening of HO length, while in vitro characterization of protein coronas provided insight into the crucial impact of HO length on opsonization-mediated immune response and elimination. The precise HO length identified in this study resulted in ESIONP probes with prolonged vascular lifetime, moderate tumor accumulation, low uptake by non-specific organs, and favorable urinary clearance, meeting prerequisites for advanced theranostic medical applications with dual MRI/PET hot spot capability and potential antitumor activity.
