Objective. Progranulin (PGRN) was recently introduced as a novel marker of chronic inflammatory response in obesity and type 2 diabetes capable of directly affecting the insulin signaling pathway. This study aimed to investigate the correlation between PGRN and type 2 diabetics with microvascular complications. Methods. PGRN serum levels and glucose metabolism related substance were measured in 84 type 2 diabetic patients with or without microangiopathies and 12 health persons. Further analyses of serum PGRN in different stages of diabetic microangiopathies were conducted. Results. Serum levels of PGRN were markedly higher in type 2 diabetic patients with microangiopathies. PGRN serum levels increased with the progress of diabetic microangiopathies with significantly highest values detectable in clinical diabetic nephropathy (CDN) and proliferative diabetic retinopathy (PDR) groups. Serum PGRN concentrations in all individuals positively and markedly correlated with systolic blood pressure (SBP), diastolic blood pressure (DBP), body mass index (BMI), triglyceride (TG), urinary albumin excretion rate (UAER), blood urea nitrogen ( BUN), creatinine (CRE), white blood cell (WBC), disease duration, IL-6, and TNF-alpha, while correlating negatively and significantly with eGFR. Multiple linear regression analysis showed that only UAER and CRE were independently associated with serum PGRN. Conclusion. PGRN might be considered as a marker for diabetic microangiopathy and its severity.

Autophagy dysregulation mediates the damage of high glucose to retinal pigment epithelium cells

Summary: The study investigated the role and mechanism of autophagy in retinal pigment epithelial cells damaged by high glucose, revealing decreased cell viability, increased apoptosis, and activated autophagic flux under high glucose conditions. These changes were reversed by treatment with an autophagy inhibitor. New strategies for managing diabetic retinopathy may be developed based on autophagy regulation.

Summary: The study found that the anti-VEGF drugs Ranibizumab and Conbercept may trigger autophagy in vascular endothelial cells, while Aflibercept may inhibit autophagy. The combination of Conbercept and autophagy inhibitors can significantly inhibit angiogenesis in vitro. The mechanism of autophagy activation is related to the activation of the p53/DRAM pathway.

Knockdown of IL4I1 Improved High Glucose-evoked Insulin Resistance in HepG2 Cells by Alleviating Inflammation and Lipotoxicity Through AHR Activation

Lin Run, Zhufang Tian, Lin Xu, Junhui Du, Nan Li, Qi Wang, Hongzhi Sun

Summary: Insulin resistance (IR) plays a significant role in the development of Type 2 diabetes mellitus (T2DM), and inflammation is closely related to IR and T2DM. This study investigated the role of Interleukin-4-induced gene 1 (IL4I1) in T2DM. The results showed that IL4I1 expression was up-regulated in peripheral blood samples of T2DM patients and high glucose-treated HepG2 cells, and silencing IL4I1 reduced inflammation, improved lipid metabolism, and attenuated IR in these cells by inhibiting the aryl hydrocarbon receptor (AHR) signaling pathway. This suggests that IL4I1 could be a potential therapeutic target for T2DM.

Summary: Nanoparticles have been successfully used in enhancing oil recovery (EOR) due to their small size and abundant surface functional groups. In this research, SiO2 nanoparticles of different sizes were synthesized and their properties in terms of interfacial tension, contact angle, viscosity, capillary pressure, and oil extraction ability were investigated. The results showed that SiO2 nanoparticles with a size of about 60nm were the most efficient oil-displacing agent for medium-permeability reservoirs.

Summary: This study investigated the expression of PLVAP in stomach adenocarcinoma (STAD) patients and its value in predicting patient prognosis. The results showed that elevated PLVAP expression was associated with advanced disease and worse clinical outcomes. Furthermore, PLVAP expression was also found to be correlated with specific bacterial infections.

Summary: Nanoparticles have been successfully applied in enhanced oil recovery due to their small size and surface functional groups. However, the effect of nanoparticle size on oil recovery has not been systematically studied. In this research, SiO2 nanoparticles of different sizes were synthesized and their properties were investigated. The experimental results showed that as the SiO2 nanoparticle size decreased, there was a slight increase in interfacial tension, oil contact angle, viscosity, and capillary pressure. Among all the sizes, the 60 nm SiO2 nanoparticle-based nanofluid showed the highest efficiency in recovering oil.
