Summary: Immune checkpoint therapy has shown breakthrough in cancer treatment, but its interaction in glioma remains controversial. In this study, we analyzed transcriptome data and conducted immunofluorescence assay to detect the features of immune checkpoints. We found that B7-H3, VISTA, and PD-L1 were associated with overall survival and grade in glioma.

Summary: Using gene differential expression and gene ontology, a set of 26 tumor suppressor genes were identified, with SASH1, STARD13, CBFA2T3, and RECK showing strong tumor suppressor effects, and EXT1, PTCH1, KLK10, and APC demonstrating weak tumor suppressor effects. These genes can be used as specific and sensitive biomarkers for diagnosis of NSCLC cancer.

Summary: High expression of TRIP13 in gliomas is associated with poor prognosis, potentially influencing tumor development by regulating co-expressed genes related to tumorigenesis and therapeutic resistance. Aberrant expression of TRIP13 may be regulated by mechanisms such as DNA methylation and miRNA targeting, leading to high aneuploidy levels and reduced CD8(+)/Treg ratio, which worsen patient outcomes.

Summary: This study aimed to investigate the histopathological characteristics of recurrent gliomas and their clinical significance. The histopathological patterns of recurrent gliomas were found to be related to survival time after reoperation. Primary WHO low-grade or IDH1 mutant gliomas showed a survival benefit mainly on later recurrence.

Summary: In this study, it was shown that circ-ITCH is significantly downregulated in glioma and can inhibit cell proliferation and invasion by regulating the miR-106a-5p/SASH1 axis, providing a new molecular target for glioma treatment.

Summary: The upregulation of TBX15 has been observed in various tumors and is known to promote uncontrolled tumor cell proliferation and inhibit apoptosis, thereby accelerating the malignant transformation of tumors. However, the prognostic significance of TBX15 in glioma and its correlation with immune infiltration are still unknown. This study aimed to investigate the prognostic value of TBX15 and its relationship with immune infiltration in glioma, as well as evaluate TBX15 expression in pan-cancer using RNAseq data.

Summary: Cancer stem cells, particularly in glioblastoma, play a crucial role in tumor resistance to treatment and recurrence. The regulatory mechanisms of glioma stem cells include tumor microenvironment, niche concept, metabolism, immunity, genetics, and epigenetics, which serve as potential targets for future treatments.Understanding the molecular markers and treatment resistance of GSCs in glioblastoma may lead to more effective therapeutic strategies in the future.

Summary: This study investigated the expression and prognostic significance of ITGB2 in glioma. The results showed that ITGB2 can be a potential marker for mesenchymal molecular subtype gliomas and an independent predictive marker of overall survival in malignant glioma patients. In addition, ITGB2 is closely related to glioma immune-related activities. Methylation of ITGB2 negatively regulates its expression in low-grade glioma tissues and can be used to predict the overall survival of low-grade glioma patients.

Summary: CD37 is abnormally expressed in diffuse gliomas and is associated with unfavorable prognosis, potentially exerting its effect by suppressing immune processes. Gliomas with high expression of CD37 have more M2 macrophage and neutrophil infiltration, with lower NK cell abundance.

Summary: To understand the metastasis of nasopharyngeal carcinoma, a genomic analysis was conducted on matched blood and tumor samples. Two distinct routes of metastasis were identified, one originating from lymph nodes and the other from primary tumors. The hematogenous route exhibited higher expression of IFN-gamma response genes and exhausted CD8(+) T cells. The hematogenous group showed better progression-free survival and PD-1 immunotherapy response, while the lymphatic group responded better to locoregional radiotherapy.

Summary: PD-L1 expression is significantly associated with low overall survival (OS) in patients with glioblastoma (GBM). However, this result needs to be interpreted with caution and further evaluated in large, multicenter clinical studies with similar baseline data.

Summary: A single-institution observational study evaluated the prognostic relevance of the methylation status of MGMT in diffuse gliomas. The study found that younger patients with surgically resected gliomas and temozolomide (TMZ) adjuvant chemotherapy had a better prognosis. The chosen method and cut-off value determination for methylation status affected the results. The study recognized MGMT promoter methylation as a positive prognostic factor, but further verification in a larger population is needed.
