Tamarixetin 3-O-β-D-Glucopyranoside from Azadirachta indica Leaves: Gastroprotective Role through Inhibition of Matrix Metalloproteinase-9 Activity in Mice

Neem (Azadirachta indica) is a well-known medicinal and insecticidal plant. Although previous studies have reported the antiulcer activity of neem leaf extract, the lead compound is still unidentified. The present study reports tamarixetin 3-O-beta-D-glucopyranoside (1) from a methanol extract of neem leaves and its gastroprotective activity in an animal model. Compound 1 showed significant protection agairist indomethacin-induced gastric ulceration in mice in a dose-dependent manner. Moreover, ex vivo and circular dichroism studies confirmed that 1 inhibited the enzyme matrix metalloproteinase-9 (MMP-9) activity with an IC50 value of ca. 50 mu M. Molecular docking and dynamics showed the binding of 1 into the pocket of the active site of MMP-9, forming a coordination complex with the catalytic zinc, thus leading to inhibition of MMP-9 activity.

Summary: The aims of this study were to investigate the phytochemical properties and chemical transformation of compounds isolated from the medicinal plant Dolichandrone atrovirens, listed in 'Ayurveda'. Several compounds were isolated from the plant, and a novel spiro-oxindolo-pyrrolizidine adduct was synthesized. The structures were determined using various analytical techniques, and the compounds were evaluated for their potential as future drug candidates using in-silico ADME studies.

Loop Dynamics and Conformational Flexibility in Dengue Serine Protease Activity: Noninvasive Perturbation by Solvent Exchange

Summary: Molecular mechanics are important for enzyme action and understanding loop motion dynamics is crucial for designing enzyme inhibitors. In this study, we investigated the structural dynamics of NS2B/NS3 dengue protease for the development of new protease inhibitors. The internal dynamics of two regions, fingers 1 and 2, adjacent to the active site triad were found to control the enzyme action. Correlated motions in the beta-turns of these regions modulated the enzyme activity. Perturbation of these motions by cosolvent concentration, temperature, and mutations affected the enzyme kinetics.

Summary: We investigated the antiulcer efficacy of 3-indolyl furanoids derived from indomethacin and found that 3g protected against indomethacin-induced gastropathy by inhibiting MMP-9. This study established a chemical modification strategy for the development of safer NSAIDs.

Naringenin-Functionalized Gold Nanoparticles and Their Role in ?-Synuclein Stabilization

Summary: In this study, the impact of a compound called naringenin on the aggregation and fibril formation of α-synuclein, which is linked to the pathology of Parkinson's disease, was investigated. The findings showed that naringenin effectively inhibits aggregation and fibril formation, thereby delaying the progression of Parkinson's disease. The compound was incorporated onto the surface of gold nanoparticles and demonstrated the ability to interact with α-synuclein and alter the absorption spectrum of the nanoparticles. Additionally, naringenin and its nanocomplex did not affect neuronal cell viability.

Solid state synthesis, thermal, spectral, optical, crystal structure and atomic packing studies of 2-(3-Hydroxyphenyl)-2,3-dihydroquinazolin-4(1H)-one

Summary: In this study, an environment friendly solid state synthesis method was used to successfully synthesize a molecular complex, 2-(3-Hydroxyphenyl)-2,3-dihydroquinazolin-4(1H)-one (ANHB), with the highest yield. The compositional behavior of composites in the entire range was determined by analyzing the phase diagram of anthranilamide (AN) 3-hydroxybenzaldehyde (HB) system, which identified the formation of two eutectics and a molecular complex. The novelty and structural behavior of ANHB were studied using various techniques such as FTIR, NMR, UV-vis, fluorescence, DSC, and PXRD. The thermodynamic properties of the eutectics and complex were characterized using the heat of fusion values obtained from DSC. The single crystal XRD (SCXRD) and atomic packing studies confirmed that ANHB crystallizes in a centro-symmetric triclinic space group P 1 with two molecules in the asymmetric unit and four molecules in the unit cell. The synthesized complex exhibited fluorescence emission in the visible region.

A novel molecular reporter for probing protein DNA recognition: An optical spectroscopic and molecular modeling study

Summary: A novel benzo[a]phenoxazine-based fluorescent dye LV2 was used to investigate the recognition of a linker histone protein H1 by calf-thymus DNA (DNA). The fluorescence lifetime and rotational relaxation time of LV2 buried in H1 or DNA showed significant changes upon the formation of the H1-DNA complex, indicating successful recognition between H1 and DNA. Molecular docking and molecular dynamics simulations demonstrated that the binding of LV2 in the interface of the H1-DNA complex was energetically more favorable. Through the significant changes in fluorescence lifetime and rotational relaxation time, the benzo[a]phenoxazine-based fluorescent dye reported on the recognition of H1 by DNA.

Single point mutations at the S129 residue of α-synuclein and their effect on structure, aggregation, and neurotoxicity

Summary: Parkinson's disease is an age-related neurological disorder caused by aggregates of alpha-synuclein (aS) proteins. In this study, the structure and aggregation behavior of two artificial single point mutants of aS were analyzed and compared to the wild-type. The mutants showed enhanced structural stability and alpha-helical propensity, resulting in slower fibril formation and reduced toxicity to neuronal cells.

Summary: Ribosomal protein S6 kinase 1 (S6K1) plays a significant role in cellular signaling pathways and is considered a potential target for drug design and discovery. This study identified Hecogenin and Glabrene as potential S6K1 inhibitors through virtual screening, molecular docking, and drug-likeliness filters. These compounds showed good affinity, specificity, and stable complexes with S6K1, suggesting their potential therapeutic use for diseases such as diabetes, obesity, and cancer.

Summary: Natural products are important for drug discovery due to their unique chemical space. Researchers have been using diversity-oriented synthesis to synthesize novel natural product hybrids and analogues. In this study, natural product hybrids of parthenin were synthesized using the weed Parthenium hysterophorus. The synthesized compounds were characterized and evaluated for their cytotoxicity, with compound 6 showing inhibitory effects on HCT-116 cells.

Experimental and Computational Insights into the Molecular Interactions between Human Transferrin and Apigenin: Implications of Natural Compounds in Targeting Neuroinflammation

This study investigates the binding and dynamics of the human transferrin (HTF)-apigenin complex, and suggests that apigenin has a neuroprotective effect by binding to HTF and maintaining a stable conformation.
