Summary: This study demonstrates that ILC2s are a non-redundant source of IL-4/IL-13 required for protective immunity against primary Nb infection. IL-4/IL-13 secretion from ILC2s is necessary for the recruitment of eosinophils to the lung and plays a crucial role in goblet cell hyperplasia and tuft cell accumulation in the small intestine.

Summary: Follicular helper CD4(+) T cells (TFH) are major sites of HIV replication, but follicular regulatory CD4(+) T cells (TFR) limit HIV replication by suppressing TFH through IL-2 restriction. TFR reduce TFH viability and HIV infected TFH, and IL-2 enhances TFH viability but decreases HIV replication in the presence of TFR.

Summary: Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by the involvement of T follicular helper (T-FH) cells. This study found that AIM2 expression was upregulated in SLE patients, especially in T-FH-like cells. Knocking out AIM2 significantly reduced T-FH cell response and decreased levels of serum autoantibodies and proteinuria in lupus mice.

Summary: We found that increased TFH cells and the expression of IL-21 contribute to TLS formation and renal fibrosis in IgA nephropathy and ischemic-reperfusion-injury-induced TLS model. Targeting the ICOS-signaling pathway could reduce TFH cell infiltration and alleviate renal fibrosis. Additionally, IL-21 stimulates renal fibrosis and the expression of p65, which can be mitigated by a p65 selective inhibitor, BAY 11-7085.

Summary: This study found that a specific subset of effector Tfh cells was activated during allogeneic kidney transplantation, and these cells played a crucial role in transplant rejection by regulating the clonal dynamics of donor-specific germinal center B cells.

Summary: CHI3L1 is found to play a significant role in allergic diseases based on data from patient cohorts and mouse models. In this study, Chi3l1 was shown to modulate T(H)2 and IL-4-producing T-FH cells, which are crucial for IgE responses.

Summary: The study showed that blocking IL-21R can inhibit the generation of Tfh cells, promote Tfr differentiation, and inhibit DSA production. In a transplant model, alpha IL-21R significantly reduced Tfh/Tfr ratio and germinal center response, leading to a significant decrease in DSA levels after transplantation, indicating a potential therapeutic role in preventing chronic antibody-mediated rejection.

Summary: During viral infection, CD4(+) T cells differentiate into Th1 cells for inflammation and pathogen clearance, or Tfh cells for enhancing germinal center reactions. The lineage-defining transcription factors T-bet and Bcl6 play key roles in determining the fate of CD4(+) T cells toward Th1 or Tfh. Transcriptional networks guided by cytokine cues dynamically shape the fate and function of these cells during infection.

Summary: The immunosuppressive microenvironment in PDAC is closely related to poor prognosis. Tfh cells promote an immunoactive tumor microenvironment by secreting CXCL13 and IL-21, correlating with better patient prognosis. However, the anti-tumor function of Tfh cells is inhibited by the PD-L1/PD-1 signaling pathway, which can be reversed by neoadjuvant chemotherapy.

Summary: The interactions between CD4(+)T cells and B cells are crucial for antibody responses and autoimmune diseases. Recent studies identified a new subset of CD4(+)B helper T cells called peripheral helper T (Tph) cells, which provide help to B cells in inflamed tissues and share some functions with Tfh cells.
