Investigations on anti-inflammatory and analgesic activities of Alnus nitida Spach (Endl). stem bark in Sprague Dawley rats

Ethnopharmacological relevance: Stem bark of Alnus nitida (Spach) Endl. (family Betulaceae) is used by local communities in northern areas of Pakistan as a remedy for swelling, injuries and pain. However no pharmacological study of this plant has been reported to confirm these activities. In this study antiinflammatory and analgesic effect of A. nitida stem bark have been evaluated. Methods: Powder of the stem bark of A. nitida was extracted with methanol (ANBM) and partitioned in escalating polarity to get the n-hexane (ANBH), chloroform (ANBC), ethyl acetate (ANBE) and the residual soluble aqueous (ANBA) fractions. The methanol extract and derived fractions were evaluated for antiinflammatory activity by using in vitro heat induced albumin denaturation assay and various in vivo assays; carrageenan-induced hind paw edema method, Freunds' complete adjuvant induced arthritis, histamine induced paw edema and xylene induced ear edema in Sprague Dawley rat. The extracts/fractions were also evaluated for analgesic effects by using hot plate analgesic test and acetic acid induced writhing test in rat. The ANBM composition was analyzed by HPLC-DAD and GC-MS analysis. Results: Results of heat induced albumin denaturation activity indicated that among the extract/fractions ANBC at concentration range of 100-500 mu g/ml remarkably protected the heat induced albumin denaturation. The pretreatment with ANBC significantly reduced the carrageenan induced edema with 90.81 +/- 1.6% after 4 h, comparing with 86.63 +/- 3.42% reduction produced by the reference drug diclofenac potassium. Histopathological alterations of the gastric and hind paw were decreased with the extract/fractions. Furthermore, anti-inflammatory effects of ANBC were evident in Freunds' complete adjuvant induced arthritis, histamine induced paw edema and xylene induced ear edema. The latency time in hot plate analgesic assay with ANBC (61.59 +/- 0.38%) after 90 min was comparable to standard drug morphine (69.31 +/- 2.67%) and aspirin (67.24 +/- 2.08%). Similarly ANBC significantly (p < 0.01) inhibited the pain sensation in acetic acid induced writhing test in rat. HPLC-DAD analysis of ANBM indicated the presence of gallic acid, catechin and rutin while the GC-MS analysis of ANBM indicated the presence of 30 compounds predominantly of neophytadiene, 3,7,11,15 tetramethyl-2-hexadecen-1-ol, phytol, vitamin E and linalool. Conclusion: The results of this study suggested that the presence of polyphenols, sterols, terpenoids and other constituents might contributed towards the anti-inflammatory and analgesic activities of the crude methanol extract of A. nitida bark and its derived fractions. This study endorsed the folklore use of A. nitida bark for inflammation related disorders.

Characterization and fabrication of zinc oxide nanoparticles by gum Acacia modesta through green chemistry and impregnation on surgical sutures to boost up the wound healing process

Summary: In this study, zinc oxide nanoparticles (ZnO-NPs) based surgical sutures were developed for accelerated wound healing. The synthesized NPs showed good antibacterial potential against Escherichia coli and Methicillin-resistant staphylococcus aureus (MRSA), and the coated suture exhibited good tensile strength. The designed suture demonstrated excellent wound healing potential in rats, with rapid epithelialization, wound contraction, mild inflammation, and absence of infection.

Synthesis, in-Silico studies, and in-Vitro bio-evaluation of new bi-thiacoumarins

Summary: In this study, two new bi-thiacoumarin derivatives, Tc01 and Tc02, were synthesized and evaluated for their antibacterial, anticancer, anticoagulant, and cytotoxic activities. Molecular docking studies revealed moderate interactions between the compounds and the target. Tc01 and Tc02 showed promising antibacterial and anticoagulant activities, with Tc01 exhibiting moderate cytotoxic activity and Tc02 showing no activity. Additionally, both compounds demonstrated significant antioxidant and antidiabetic activities.

Antioxidant and Pulmonary Protective Potential of Fraxinus xanthoxyloides Bark Extract against CCl4-Induced Toxicity in Rats

Summary: This study aimed to evaluate the antioxidant and pulmonary protective potential of Fraxinus xanthoxyloides bark extract and its fractions. The highest total phenolics, flavonoids, and terpenoids contents were found in the methanol bark extract, ethyl acetate fraction, and chloroform fraction, respectively. The most potent antioxidant capacity was shown by the methanol bark extract and ethyl acetate fraction. The administration of carbon tetrachloride induced oxidative stress and lung tissue damage, but treatment with the bark extract restored these parameters to normal levels and protected the lung tissues.

Effects of Phoenix dactylifera against Streptozotocin-Aluminium Chloride Induced Alzheimer's Rats and Their In Silico Study

Exploration of polyacrylamide microplastics and evaluation of their toxicity on multiple parameters of Oreochromis niloticus

Summary: Polyacrylamide microplastics produced in the environment have adverse effects on aquatic organisms, causing global concern. This study aimed to assess the toxicity of different concentrations of polyacrylamide microplastics on Oreochromis niloticus. Characterization of the microplastics was conducted using Fourier transform infrared and Raman spectroscopy, confirming their presence in the gills of the fish. The microplastics were found to significantly affect antioxidant enzymes, increase oxidative stress levels, and induce histological changes in various organs, highlighting their harmful effects on fish health.

Astragalus roseocephalus, a new species of section Poliothrix from Pakistan

Summary: This article describes and illustrates a new plant species, Astragalus roseocephalus sp. nov., found in Islamabad Capital Territory (Margalla hills), Rawalpindi district, Hazara, and Kashmir. It belongs to the Astragalus section Poliothrix Bunge and is closely related to Astragalus leucocephalus Benth. Morphologically, it differs from A. leucocephalus in various aspects such as height, stipule shape, leaf length, leaflet size, peduncle length, corolla color, standard size and shape, fruit shape, and number of seeds. The article also discusses the anatomy, pollen morphology, distribution, and conservation status of the new species.

Adenoid Cystic Carcinoma of the Breast: A Case Report

Summary: Adenoid cystic carcinoma (ACC) is a rare subtype of breast cancer with distinct histologic patterns. This report presents a rare case of ACC with a cribriform, trabecular growth pattern. While imaging studies are useful for screening and planning, they may not provide specific information. Pathological examination remains the standard reference for diagnosis and treatment decisions. Surgery is currently the primary therapy for breast ACC patients, but there is no clear consensus on the selection of specific surgical treatments.

Ameliorative effect of scopolamine-induced cognitive dysfunction by Fufangmuniziqi formula: The roles of alkaloids, saponins, and flavonoids

Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

The results showed that M. longifolia extract is rich in phenolic compounds and has no observed toxicity up to a certain dosage limit.

A study integrated metabolomics and network pharmacology to investigate the effects of Shicao in alleviating acute liver injury

Network pharmacology was used to construct a network of metabolite-reaction-enzyme-gene constituents, and key components and hub genes were screened. Shicao showed hepatoprotective effects against acute liver injury in rats.
