Summary: This study aimed to optimize the high pressure homogenization (HPH) process conditions and emulsifier formulation to produce nanostructured lipid carriers (NLC). The results showed that using SL, Tween 80, and WPI as emulsifiers, with a pressure of 700 bar and 2 cycles, NLC in the nanometric range could be produced and remained stable during 15 days of storage at 25°C.

Summary: The challenge faced by researchers in achieving sufficient ocular bioavailability is due to the complex structure of the eye and its protective physiological mechanisms. Low viscosity eye drops and short ocular residence time contribute to low drug concentration at the target site. Solid lipid nanoparticles (SLNs) and nanostructured lipid carriers (NLCs) are being developed as drug delivery platforms to overcome these challenges and enhance ocular bioavailability. They are biocompatible, biodegradable, and can be sterilized and scaled-up. Surface modifications can prolong ocular residence time, enhance penetration, and improve performance.

Summary: The present study aimed to improve the oral bioavailability and prokinetic activity of levosulpiride (LSP) by developing and optimizing LSP-loaded nanostructured lipid carriers (LSP-NLCs). The optimized LSP-NLCs formulation showed improved physicochemical properties and solid-state characterization. In vivo pharmacokinetic evaluation in rats revealed significantly increased AUC and C-max for LSP-NLCs compared to LSP dispersion. Additionally, LSP-NLCs demonstrated enhanced gastric emptying, intestinal transit, and fecal count. These findings suggest that LSP-NLCs could be a promising strategy for the efficient treatment of gastric diseases.

It discusses issues such as incorporation and release of bioactives, oxidative stability, lipid digestion and absorption, intestinal transport, and encapsulation and delivery of nutraceuticals. The review also highlights the performance of these nanoparticles in improving stability, bioavailability, and bioactivities of curcuminoids.

The PZ-loaded NLCs exhibited small particle size, negative zeta potential, and high drug entrapment efficiency and loading capacity. In vitro and in vivo studies confirmed the enhanced bioavailability of pioglitazone when formulated as PZ-loaded NLCs.

Summary: The ability of mesenchymal stromal cells (MSCs) to release immunomodulatory factors makes them promising candidates for treating inflammatory bowel diseases (IBD). However, nude MSC administration has limitations such as loss of immunomodulatory phenotype and low persistence. In this study, a licensing hydrogel system was designed to overcome these limitations and achieve continuous delivery of bioactive factors. The results showed that encapsulated MSCs in the hydrogel limited the progression of IBD in mice.

Summary: Non-alcoholic fatty liver disease (NAFLD) affects a significant portion of the adult population worldwide and no approved pharmacologic therapy is currently available. A novel drug delivery system called lipid nanocapsules (LNCs) has shown promise in treating NAFLD by inducing the secretion of glucagon-like peptide 1 (GLP-1). This study aimed to compare the efficacy of the LNCs system with subcutaneous injection of a GLP-1 analog in two mouse models of NAFLD. The results demonstrated that chronic administration of the LNCs system improved glucose homeostasis and insulin resistance, and prevented disease progression to a greater extent than subcutaneous injection in both models.
