Summary: The absence of Isc1 in yeast leads to mitochondrial dysfunction, but this defect can be rescued by the deletion of phospholipase Pgc1. Pgc1 regulates lipid homeostasis by inhibiting the activities of other phospholipases and controlling phospholipid synthesis. The interconnection of Isc1 and Pgc1 plays a significant role in maintaining lipid balance in the cell.

Summary: This study demonstrates the superior performance of chemical vapour deposition (CVD) coating for suppressing the electroosmotic flow (EOF) compared to liquid phase deposition (LPD) in capillary electrophoresis (CE). By adjusting the modification method and surface chemistry, the capillary can be easily and effectively modified, and the stability of the coating meets the long-term monitoring requirements for water chemistry.

Summary: This study reveals that the metabolic fate of newly synthesized phosphatidic acid (PA), the lipid precursor of phosphatidylinositol (PI), is influenced by the fatty acyl-CoA used, leading to preferential routing of the arachidonoyl-enriched species towards PI synthesis. Additionally, the unsaturated forms of PI(4,5P)(2) are replenished faster than the saturated forms during PLC activation, indicating a favored recycling of the unsaturated forms of the PLC-generated hydrolytic products. CDS2 contributes to increased PI resynthesis during PLC activation, while Nir2 contributes to rapid PPIn resynthesis, but the faster re-synthesis of the 38:4 species does not depend on Nir2. Therefore, the fatty acid side-chain composition of the lipid precursors used for PI synthesis is an important determinant of their metabolic fates and the maintenance of the unique fatty acid profile of PPIn lipids.

Summary: Cells sense extracellular signals and regulate their metabolism and physiology accordingly. The plasma membrane plays a crucial role in this process by acting as an informational barrier and responding to regulatory inputs. Phosphatidic acid (PA) is a well-known intracellular messenger that regulates various cellular processes and is involved in plant growth, development, and stress tolerance. This review focuses on the primary signaling events triggered by rapid PA accumulation in plant cells, and discusses its role in mediating the response to hormones and hormone-like regulators.

Summary: Capillary electrophoresis with fluorescence detection (CE-F) is a powerful method for measuring enzyme activation in single cells. However, the reporter substrates used in cellular enzymatic assays for CE-F may affect enzyme kinetics. To address this issue, a fix and click method was developed to attach a fluorophore to an enzyme activation reporter prior to analysis by CE-F, allowing for the investigation of sphingolipid signaling in single cells.

Summary: Phospholipase C (PLC) metabolizes phosphatidylinositol 4,5-bisphosphate in the plasma membrane and generates inositol 1,4,5-trisphosphate and diacylglycerol, which regulate cellular functions. Mammalian PLC consists of six typical classes and one atypical class, and dysregulation of PLC activity is associated with various diseases. Therefore, the review focuses on the structure, activation mechanisms, and physiological functions of mammalian PLC.

Summary: In this study, a quadruple UV LED array was designed to directly irradiate gel electrophoresis chips at 275 nm. Simple detection setup using a 365 nm filter, UV camera lens, and CCD was integrated with the small gel electrophoresis chip to construct the IFI-GEC device. The LED array exhibited good evenness and enabled online imaging of the entire gel electrophoresis chip. The reproducible IFI-GEC detection was completed within 10 minutes with a low LOD of 40 ng for lysozyme detection, demonstrating the potential of the IFI-GEC device for stain-free online and portable protein detection.

Summary: This study presents the crystal structure of a fungal NPC derived from Rasamsonia emersonii, revealing its distinguishable features from other known phospholipase C. The structure of the fungal NPC lacks the C-terminal domain (CTD) found in plant NPCs, which is crucial for their activity. Through comparative analysis and mutation studies, a potential molecular mechanism underlying phospholipid hydrolysis in fungal NPCs is proposed. Furthermore, the study sheds light on the evolutionary trajectory of enzymes across diverse species.

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.
