Summary: The polyphenolic compound SDG found in flaxseed has been shown to protect mice from BaP-induced liver and kidney toxicity by regulating oxidative stress, inflammation, and apoptosis. SDG administration resulted in lower levels of liver and kidney damage markers and modulation of specific signaling pathways.

Summary: miR-30a plays an important role in the tumor suppressor gene of trophoblastic cells, but its role in hydatidiform moles is unclear. This study found that downregulation of miR-30a promotes proliferation, migration, and invasion of trophoblastic cells, and activates the ERK, AKT, and P38 signaling pathways by regulating STOX2. These results provide new insights into the role of miR-30a in regulating trophoblastic cell transformation.

Summary: The combination of everolimus and bortezomib has shown synergistic antiproliferative and apoptotic effects against fibrosarcoma and osteosarcoma, suppressing pulmonary metastases. Everolimus and bortezomib inhibit tumor growth via the JNK/p38/ERK MAPK and AKT pathways. This combination therapy shows promise as a new therapeutic strategy for bone and soft tissue sarcomas.

Summary: Beta-elemene regulates the balance of proinflammatory cytokines in mouse white adipose tissue through MAPK signaling, affecting the imbalance of M1-M2 macrophages and inhibiting the phosphorylation of MAPK pathways. These results suggest that beta-elemene may be a potential therapeutic medicine for macrophage-mediated chronic diseases.

Summary: Fibrosis is a crucial factor contributing to cardiac dysfunction after myocardial infarction (MI). This study investigated the function of a protein called OTUD7B in cardiac fibrosis and identified its underlying mechanism. The researchers found that OTUD7B expression is downregulated in a MI rat model and in cultured cardiac fibroblasts under hypoxic conditions and TGF-beta 1 treatment. They also discovered that OTUD7B regulates the phosphorylation of FAK and ERK/P38 MAPK, leading to decreased expression of alpha-SMA and collagen I, both markers of fibrosis. This suggests that OTUD7B could be a potential therapeutic target for fibrosis after MI.

Summary: The up-regulation of RNF2 in liver fibrosis enhances HSC activation and exerts fibrogenic effects by inhibiting the ERK/p38 signaling pathway induced by TGF-beta 1. The inhibition of RNF2 may be a promising therapeutic target for liver fibrosis.

Summary: This study investigated the inhibitory effect of Yohimbine hydrochloride (YH) on melanogenesis and revealed its underlying molecular mechanism. The results showed that YH effectively suppressed melanin production by inhibiting the Wnt/β-catenin and p38/MAPK signaling pathways. Thus, YH may have potential as a skin-whitening compound for preventing hyperpigmentation disorders in the future.

Summary: The study focused on the effects of altersolanol B (AB), a minor fungal metabolite, on ER+ breast cancer cells. AB demonstrated antiproliferative activity and induced apoptosis in these cells. It also disrupted AKT and ERK1/2 signaling pathways and inhibited NF-kappa B activation. The findings support further investigation of AB and similar compounds as potential anticancer drugs.

Summary: This study comprehensively characterized a panel of patient-derived BRAFV600E positive melanoma cell lines and found that a decrease in sensitivity to MAPK/ERK pathway inhibition corresponded with increased migratory potential and the acquisition of stem cell-like properties. Additionally, it was discovered that increased activity of the PI3K/AKT pathway lead to a decreased dependency on MAPK/ERK signaling in melanoma cells.

Summary: miR-133a is strongly associated with GIO and similar disorders induced by glucocorticoids in MSCs. Silencing miR-133a resulted in positive effects on GC-treated MSCs and on bone loss in GIO animal models. Moreover, the FGFR1-MAPK/ERK signaling may be involved in the protective effect of miR-133a silencing.
