Summary: This study establishes the role of interleukin 26 (IL-26) in the generation of IL-17A producing Th17 CD4(+) T cells, indicating its involvement in the pathogenic response of psoriasis. IL-26-producing cells infiltrate psoriatic skin and promote their own differentiation into IL17A-producing T(H)17 cells through epithelial crosstalk involving paracrine production of TGF-& beta;1.

Summary: Mesenchymal stromal cells (MSC) are potential cellular therapeutics for psoriasis, a chronic inflammatory skin and joint disease. MSC adoptive cellular therapy can modulate the immune response and suppress the hyperactivation of effector T cells underlying the disease. In this study, we investigated the therapeutic potential of bone marrow and adipose tissue-derived MSCs in a psoriasis-like inflammation model. We found that both unlicensed and licensed MSCs accelerated the healing of psoriatic lesions, but unlicensed MSCs were more efficient in resolving skin inflammation.

Summary: The study identified oncologic miR-182 as a new regulator of pulmonary artery hypertension (PAH) targeting Myadm, and suggested that the balance between the BMP and TGF-beta signalling pathways is disrupted in PAH. Hypoxia triggers pathological progression in PAH by downregulating BMP/SMAD1/5/8 expression and enhancing TGF-beta/SMAD2/3 signalling pathway. miR-182 gain-of-function inhibits the progression in PAH, while loss-of-function increases the pathological progression. The study also indicated a crosstalk between endothelial cells and smooth muscle cells in the BMP-TGF-beta pathway associated with NOS3/NO/cGMP.

Summary: After allergen challenge, IL-6 trans-signalling proteins showed increased activity in the airways, potentially leading to augmented inflammation and tissue damage. Different cell types expressed IL-6R subunits and could contribute to the production of sIL-6R, while bronchial fibroblasts activated by IL-6 trans-signalling produced increased levels of chemokines. These findings suggest a role for IL-6 trans-signalling in exacerbating inflammation and lung dysfunction in asthma.
