Toxicity of long chain fatty acids towards acetate conversion by Methanosaeta concilii and Methanosarcina mazei

Long-chain fatty acids (LCFA) can inhibit methane production by methanogenic archaea. The effect of oleate and palmitate on pure cultures of Methanosaeta concilii and Methanosarcina mazei was assessed by comparing methane production rates from acetate before and after LCFA addition. For both methanogens, a sharp decrease in methane production (>50%) was observed at 0.5mmolL(-1) oleate, and no methane was formed at concentrations higher than 2mmolL(-1) oleate. Palmitate was less inhibitory than oleate, and M.concilii was more tolerant to palmitate than M.mazei, with 2mmolL(-1) palmitate causing 11% and 64% methanogenic inhibition respectively. This study indicates that M.concilii and M.mazei tolerate LCFA concentrations similar to those previously described for hydrogenotrophic methanogens. In particular, the robustness of M.concilii might contribute to the observed prevalence of Methanosaeta species in anaerobic bioreactors used to treat LCFA-rich wastewater.

Overflow metabolism at the thermodynamic limit of life: How carboxydotrophic acetogens mitigate carbon monoxide toxicity

Summary: Carboxydotrophic metabolism is gaining interest for its applications in gas fermentation technology, where carbon monoxide is converted into fuels and commodities. Acetogenic carboxydotrophs, such as C. autoethanogenum, play a crucial role in this process. To cope with the toxicity of CO, these organisms employ a form of overflow metabolism that allows them to efficiently use CO at varying substrate influxes. The study focuses on how C. autoethanogenum utilizes acetate reduction to ethanol as a form of overflow metabolism when grown solely on CO and syngas mixtures.

Microbial conversion of carbon dioxide and hydrogen into the fine chemicals hydroxyectoine and ectoine

Summary: This study explores a novel conversion of CO2 into high-value chemicals hydroxyectoine and ectoine in the pharmaceutical industry. Through literature search and genomic mining, 11 species of microbes capable of using CO2 and H-2 with the genes for ectoine synthesis were identified. Laboratory tests confirmed that Hydrogenovibrio marinus, Rhodococcus opacus, and Hydrogenibacillus schlegelii are the most promising bacteria for CO2 to ectoines bioconversion. These results provide the first proof of a novel valorization platform for CO2 and lay the foundation for a new economic niche in CO2 recircularization into pharmaceuticals.

Sialylation and Sulfation of Anionic Glycoconjugates Are Common in the Extracellular Polymeric Substances of Both Aerobic and Anaerobic Granular Sludges

Summary: This study investigates the sialylation and sulfation of anionic glycoconjugates in anaerobic and aerobic granular sludge. The results showed that these two glycoconjugate modifications commonly occur in the extracellular polymericsubstances (EPS) of both types of sludge. Mass spectrometry analysis and gene identification provided insights into the distribution and synthesis of these modifications in the sludge communities.

A Review of Computational Modeling in Wastewater Treatment Processes

Summary: Wastewater treatment companies are facing challenges in energy efficiency optimization, meeting stricter water quality standards, and resource recovery potential. Computational models have been recognized as effective tools for addressing these challenges and improving economic and operational efficiencies of wastewater treatment plants. Deterministic, stochastic, and time series-based models have been developed to predict the performance of wastewater treatment plants. Mechanistic models, although dominant, may introduce model structure uncertainty and require frequent calibration. Data-driven models, which search for relationships in available data, are becoming more attractive with the increasing amount of data. This review discusses the implementation of machine learning models for predicting WWTP effluent characteristics, wastewater inflows, anomaly detection, and energy consumption optimization. Hybrid models, merging mechanistic and machine learning models, are presented as a promising approach. The review also assesses gaps and future directions in the implementation of mathematical modeling in wastewater treatment processes, focusing on explainability of data-driven models and the use of Transfer Learning processes.

Design and thermodynamic analysis of a pathway enabling anaerobic production of poly-3-hydroxybutyrate in Escherichia coli

Summary: This study proposes a novel metabolic engineering strategy to achieve efficient anaerobic production of poly-(R)-3-hydroxybutyrate (PHB) in Escherichia coli. The strategy involves rerouting the glycolytic flux through non-oxidative glycolysis and coupling PHB synthesis with NADH re-oxidation. Stoichiometric analysis and various thermodynamic approaches were used to assess the feasibility and bottlenecks of the engineered pathway.

Effect of Endogenous Methane Production: A Step Forward in the Validation of Biochemical Methane Potential (BMP) Tests

Summary: This study evaluates the impact of inoculum type, pre-consumption of residual substrate, and the ratio of headspace volume to working volume on Biochemical Methane Potential (BMP) measurements. The findings suggest that degassing and discounting methane produced in blanks are essential for accurate results. The residual substrate concentration of different inocula also plays a role in the determination.

These tools are user-friendly and valuable resources for interpreting microbial activities in nature and biotechnological processes.
