Summary: The research found that allele-specific DNA methylation within the TERT promoter plays a significant regulatory role in cancer, especially more common in adult cancers and enriched in tumors with TERT promoter mutations. This allele-specific DNA methylation is essential for the regulation of TERT expression in cancer.

Summary: TLX2 plays an important role in multiple tumors and its abnormal expression is associated with a poor overall survival rate in several cancer types. The most common type of alteration in TLX2 is amplification, and it co-occurs with alterations in other genes. TLX2 has high methylation levels in various tumors. Its expression is related to immune infiltration, immune checkpoint genes, and chemoresistance.

Summary: In this study, the research group identified a list of 83 genes potentially modulated by DNA methylation in GC cell lines, with LRRC37A2 playing a role in undifferentiated and poorly differentiated tumors. The study showed that reduced LRRC37A2 mRNA levels and increased LRRC37A2 exon methylation are associated with GC progression, emphasizing the potential utility of gene-related stratification for epigenetic therapy in GC patients.

Summary: The abnormal expression of NR2F1-AS1 in gastric cancer is related to its correlation with NR2F1 expression, as well as their response to epithelial-to-mesenchymal transition signaling. NR2F1-AS1 modulates the miR-29a-3p/VAMP7 axis and plays a crucial role in the progression of gastric cancer.

Summary: Comprehensive molecular studies were conducted on thyroid tumors to identify prognostic biomarkers. Telomere-related alterations, especially TERT and TERC, were found in aggressive tumors. Short telomeres led to reorganization in the 5p subtelomeric region, resulting in increased gene upregulation and a more relaxed chromatin configuration in the tumors.

Summary: This study aimed to investigate the role of LINC00467 in the growth and metastasis of gastric cancer (GC). The results showed that LINC00467 promoted the methylation of the Reprimo promoter by recruiting DNMT1, leading to decreased expression of Reprimo. Knockdown of LINC00467 weakened the malignant properties of GC cells and reduced tumorigenesis and metastasis in vivo.

Summary: The study aimed to investigate the association between ZNF582 gene methylation and cervical cancer, as well as its correlation with high risk HPV16/18 infection. The findings showed that ZNF582 gene methylation had high specificity and sensitivity in detecting highly diseased cervical lesions, and ZNF582 protein was highly expressed in cervical cancer tissues but with low mRNA expression.

Summary: Protein and DNA methylation play important roles in various biological functions. Abnormal regulation of methyltransferases has been implicated in cancer, but its association with autophagy and carcinogenesis in breast and lung cancer is not well understood. In this study, we used UALCAN to analyze breast and lung cancer databases and found that 17 methyltransferases are upregulated in these cancers. We investigated the effects of methylation inhibition on breast and lung cancer cell lines and found that inhibiting methylation reduced cell migration, growth, and autophagy activity. This suggests that targeting methylation ability could be a potential treatment for breast and lung cancer.

Summary: DNA methylation signatures can be used as reliable biomarkers to predict patient prognosis and immune status in tumors. This study introduces the concept of tumor-based expression quantitative trait methylation (eQTM) for the identification of predictive biomarkers. eQTM CpGs and genes represent new and efficient candidate targets for prognosis and immune status monitoring.

Summary: This study reveals the significance of PCIF1 and m(6)Am modification in gastric cancer. Increased expression of PCIF1 and m(6)Am modification are associated with gastric cancer progression and poor prognosis. Silencing PCIF1 inhibits the proliferation and invasion of gastric cancer cells, and suppresses tumor growth and metastasis in a mouse model. PCIF1 modifies TM9SF1 mRNA with m(6)Am, leading to decreased TM9SF1 translation. TM9SF1 reverses the effects of PCIF1 on gastric cancer cell aggressiveness.

Summary: This study reveals that DNA methylation can drive tissue-specific enhancer basal transcription and target gene expression in human cancers. By integrating methylome, transcriptome, and 3D genomic data, enhancer methylation triplets were identified and regulatory patterns within them were dissected. Cancer-specific core TFs regulated by enhancers were observed to shape enhancer methylation forming enhancer methylation-driven CRCs (emCRCs).

Summary: This study evaluated the prognostic value of Early B-cell factors (EBFs) in gastric cancer (GC), identifying that increased EBFs were associated with poor prognosis. A prognostic model was established and validated in different cohorts, with impacts on DNA methylation, signaling pathways, and potential treatments such as immunotherapy and chemotherapy targeting EBFs.

Summary: Gastric cancer is a major cause of cancer mortality, and abnormal DNA methylation in gene promoter regions plays a key role in the development of gastric cancer. Helicobacter pylori infection is identified to be associated with gastric cancer. Abnormal DNA methylation triggers inactivation of multiple genes, and is more common in cancer-related processes than mutations. The extent of abnormal DNA methylation is closely linked to the risk of developing gastric cancer.
