Schistosomiasis is a parasitic disease caused by several species of trematode worms and it is believed that more than 261 million people are affected worldwide. New drug development has become essential because there is a risk of the parasite becoming resistant to Praziquantel, the only drug available for this infection. This study evaluated parasitological, immunological and histological parameters in mice infected with Schistosoma mansoni and treated with an herbal commercial medicine. This drug consists of menthol (30-55%) and menthone (14-32%). A 60 day treatment regimen with the herbal medicine decreased the number of S. mansoni eggs in the feces, liver, and intestine and reduced the number of hepatic granulomas. We observed a reduction of 84% in blood eosinophilia and a decrease in the IL-4 and IL-10 blood levels after treatment. Therefore, we propose that schistosomiasis treatment with this herbal medicine for 60 days has an immunomodulatory and anti-inflammatory action in this animal model for schistosomiasis thus contributing to the decrease in physio pathological effects caused by S. mansoni infection.

β2 Integrin-Mediated Susceptibility to Paracoccidioides brasiliensis Experimental Infection in Mice

Stephan Alberto Machado de Oliveira, Janayna Nunes Reis, Elisa Catao, Andre Correa Amaral, Ana Camila Oliveira Souza, Alice Melo Ribeiro, Lucia Helena Faccioli, Fabiana Pirani Carneiro, Clara Luna Freitas Marina, Pedro Henrique Burgel, Larissa Fernandes, Aldo Henrique Tavares, Anamelia Lorenzetti Bocca

Summary: The results of the study indicate that CD18(low) mice infected with Pb18 survived with fewer granulomas, lower fungal burden, lower nitrate levels, and high IgG1 levels compared to WT mice. In addition, the in vitro macrophages from CD18(low) mice showed slower internalization of yeast cells and lower fungal burden, suggesting that beta 2 integrins play a crucial role in fungal survival within macrophages.

NLRC4 inhibits NLRP3 inflammasome and abrogates effective antifungal CD8+ T cell responses

Camila O. S. Souza, Natalia Ketelut-Carneiro, Cristiane M. Milanezi, Lucia H. Faccioli, Luiz G. Gardinassi, Joao S. Silva

Summary: The NLR molecule NLRC4 has been found to promote susceptibility to experimental PCM by regulating cytokine production and protective effector mechanisms. It suppresses prostaglandin E-2 production early in infection, reducing IL-1 beta release and impacting IL-18 release later in the disease, ultimately affecting immune responses and mortality.

Schistosomicidal evaluation of synthesized bromo and nitro chalcone derivatives

Camila Marcovicz, Guilherme dos Anjos Camargo, Bianca Scharr, Larissa Sens, Mariana Negrelli Levandowski, Thiago De Castro Rozada, Patricia Castellen, Juliana Inaba, Rosimeire Nunes De Oliveira, Julio Cesar Mine, Sheila de Andrade Penteado Correa, Silmara Marques Allegretti, Barbara Celnia Fiorin

Summary: This study synthesized Chalcones with nitro and bromo substituents in different positions and investigated their conformation and schistosomicidal effects. The results showed that Chalcones had a preference for s-cis geometry and exhibited significant effects on adult Schistosoma mansoni. Chalcones reduced the motor activity, caused mortality, and suppressed egg production in a concentration-dependent manner. These findings indicate the potential of Chalcones as schistosomicidal drugs and contribute to the understanding of the relationship between molecular conformation and biological effects.

Metabolic Profile in Plasma AND CSF of LEVODOPA-induced Dyskinesia in Parkinson's Disease: Focus on Neuroinflammation

Bruno L. Santos-Lobato, Luiz Gustavo Gardinassi, Mariza Bortolanza, Ana Paula Ferranti Peti, Angela V. Pimentel, Lucia Helena Faccioli, Elaine A. Del-Bel, Vitor Tumas

Summary: Metabolomic analysis revealed a distinct metabolic profile associated with levodopa-induced dyskinesia (LID) in Parkinson's disease (PD), particularly characterized by dysregulation of the glycosphingolipid metabolic pathway and bile acid biosynthesis metabolites in plasma and CSF. These findings suggest a potential link between lipid metabolism dysregulation and LID in PD.

Metabolic alterations in Strongyloidiasis stool samples unveil potential biomarkers of infection

Allan Daniel Ribeiro Sotelo Montanhaur, Estela de Oliveira Lima, Jeany Delafiori, Cibele Zanardi Esteves, Cesar Corat Ribeiro Prado, Silmara Marques Allegretti, Marlene Tiduko Ueta, Carlos Emilio Levy, Rodrigo Ramos Catharino

Summary: Strongyloidiasis, caused by Strongyloides stercoralis infection, is a common parasitic disease in tropical or subtropical areas. Due to the lack of a gold-standard diagnostic tool, this study used metabolomics to evaluate metabolic changes in stool samples and identified 5 potential biomarkers, indicating the involvement of microbiota remodeling, parasite constitution, and host response in the intestinal metabolism of S. stercoralis infection.

The effect of Spondias mombin L. against Strongyloides venezuelensis: An in vitro approach

Paula Berna da Silva Medeiros, Tiago Manuel Fernandes Mendes, Vera Lucia Garcia, Rosimeire Nunes De Oliveira, Leticia Bastos Duart, Silmara Marques Allegretti

Summary: In this study, the ethanolic extract of Spondias mombin L. was tested for its in vitro effect against Strongyloides venezuelensis. The results showed that the extract and its fractions had a better in vitro effect than ivermectin. Fraction 4 exhibited the best results, causing 100% mortality in a short period of time. Scanning electron microscopy and gas chromatography analysis provided insights into the mechanism of action, suggesting that monoaromatic phenolic lipids may be responsible for the antiStrongyloides effect. The use of polyvinylpyrrolidone reduced the efficiency of the extract.

Summary: The study evaluated the levels of plasma soluble HLA-G (sHLA-G) in COVID-19 patients and healthy controls. The results showed increased levels of sHLA-G in COVID-19 patients, which correlated with disease severity. However, the levels decreased in critically ill patients, possibly indicating immune exhaustion. sHLA-G also exhibited positive correlation with other disease mediators IL-6, IL-8, and IL-10.

Summary: SARS-CoV-2 infection leads to distinct disease patterns with significant changes in host regulatory responses. Severe cases suffer from lung inflammation and systemic effects, while critically ill patients experience a lipid storm that affects inflammation and tissue damage. Sphingolipids play critical roles in various cellular processes and may serve as potential biomarkers and therapeutic targets for COVID-19.
