Summary: Reduced STAT3 activity in mice leads to increased susceptibility to DSS-induced colitis, indicating that the IL-6/gp130/STAT3 signaling pathway orchestrates intestinal barrier function by modulating cytokine secretion and promoting epithelial integrity to maintain defense against bacteria. Therapeutically targeting this signaling axis may serve as a treatment strategy for IBD patients.

Summary: Rv2145c protein stimulates production of IL-10 and other cytokines in macrophages, enhancing bacterial growth and delaying clearance in the lungs and spleens of mice through modulation of host signals. Antibody treatment against IL-10 receptor restores the clearance of Rv2145c-expressing bacteria, suggesting a role for Rv2145c in creating a favorable environment for bacterial survival.

Summary: Aberrant amino acid metabolism, particularly the excessive plasma kynurenine (Kyn), is common in obesity. This study demonstrates that overexpressed indoleamine 2,3-dioxygenase 1 (IDO1) in adipocytes is the primary source of excessive Kyn, highlighting the central role of adipocytes in Kyn metabolism. Depletion of Ido1 in adipocytes eliminates Kyn accumulation, protecting mice against obesity.

Summary: Gene expression in individual cells is regulated by various epigenetic mechanisms, but the predictive ability of epigenomic analysis for single cells is limited. A new method proposed in this study allows for the analysis of multiple components of the epigenome in the same single cell, enabling reanalysis and identification of reproduced reads. This method provides mechanistic insights for cellular phenotypes through multilayered epigenome analysis, with the ability to predict active signaling pathways in single cells.

Summary: This study demonstrates that NOD1 functions as a major regulator in hematopoietic cells, affecting the generation and differentiation of lymphoid progenitors and peripheral T lymphocyte homeostasis. NOD1 mediates these functions by facilitating STAT5 signaling. Loss of functional NOD1 leads to a decrease in mature T, B, and natural killer cells, and impairs anti-tumor immunity and prevents colitis.
