Romanian wild boars and Mangalitza pigs have a European ancestry and harbour genetic signatures compatible with past population bottlenecks

We aimed to analyse the genetic diversity of Romanian wild boars and to compare it with that from other wild boar and pig populations from Europe and Asia. Partial sequencing of the mitochondrial encoded cytochrome b (MT-CYB) gene from 36 Romanian wild boars and 36 domestic pigs (Mangalitza, Bazna and Vietnamese breeds) showed that the diversity of Romanian wild boars and Mangalitza pigs is fairly reduced, and that most of the members of these two populations share a common MT-CYB haplotype. Besides, in strong contrast with the Bazna animals, Romanian wild boars and Mangalitza swine did not carry Asian variants at the MT-CYB locus. The autosomal genotyping of 18 Romanian wild boars with the Illumina Porcine SNP60 BeadChip revealed that their genetic background is fundamentally European, even though signs of a potential Near Eastern ancestry (similar to 25%) were detectable at K = 4 (the most significant number of clusters), but not at higher K-values. Admixture analysis also showed that two wild boars are of a hybrid origin, which could be explained by the mating of feral animals with domestic pigs. Finally, a number of Romanian wild boars displayed long runs of homozygosity, an observation that is consistent with the occurrence of past population bottlenecks and the raise of inbreeding possibly due to overhunting or to the outbreak of infectious diseases.

Characterizing the Mitochondrial Diversity of Arbi Goats from Tunisia

Transversal gene expression panel to evaluate intestinal health in broiler chickens in different challenging conditions

Eimeria challenge altered expression of genes related to barrier function and metabolism, increasing expression of genes related to immune response and oxidative stress. Detecting the footprint of selection on the genomes of Murciano-Granadina goats

Summary: Artificial selection plays a major role in altering the genetic composition of livestock populations, with identifying genes under selection providing insight into their effects on phenotypic variation. A study on Murciano-Granadina goats identified 77 candidate genomic regions/SNPs, with significant selective sweeps on multiple chromosomes. RNA-Seq data revealed differential expression of 6414 genes during lactation, with 183 genes overlapping with selective sweeps related to various metabolic and developmental functions. The MC1R gene polymorphism was found to be the main determinant of black and brown colorations in these goats.

Supplementation of Estradiol Into Semen Extender Improved Goat Sperm Cryopreservation

Summary: The study found that supplementation of 3 and 5 mu M concentrations of estradiol can improve sperm viability and membrane integrity in goat semen, with the 3 mu M concentration also increasing the highest rate of progressive motility. However, a higher concentration of estradiol (5 mu M) reduced progressive motility. Additionally, there is a considerable interaction between EDTA and estradiol for sperm motility, with EDTA reducing the supportive effects of estradiol on sperm cryopreservation parameters.

An Electrochemical Protocol for CRISPR-Mediated Gene-Editing of Sheep Embryonic Fibroblast Cells

Summary: In this study, a protocol for electrotransfection of sheep embryonic fibroblast cells was developed. Optimal pulsing conditions and the addition of 3% dimethyl sulfoxide (DMSO) resulted in high transfection efficiency (>98%) and cell survival rate (>97%). CRISPR/Cas9-mediated gene editing of BMP15 and GDF9 genes in sheep EF cells was successfully achieved using the electroporation method.

Variability in porcine microRNA genes and its association with mRNA expression and lipid phenotypes

Summary: Our study identified SNPs in miRNA genes and their binding sites in porcine 3' UTR, and showed a reduced variability of porcine miRNA genes, suggesting strong purifying selection, particularly in the seed region. We also found significant associations between specific miRNA SNPs and lipid-related traits and hepatic gene expression in pigs, highlighting the importance of investigating the functional effects of miRNA polymorphisms outside the seed region.
