Cucurbitacin D exhibits potent anti-cancer activity in cervical cancer

In this study, we for the first time, investigated the potential anti-cancer effects of a novel analogue of cucurbitacin (Cucurbitacin D) against cervical cancer in vitro and in vivo. Cucurbitacin D inhibited viability and growth of cervical cancer cells (CaSki and SiHa) in a dose-dependent manner. IC50 of Cucurbitacin D was recorded at 400 nM and 250 nM in CaSki and SiHa cells, respectively. Induction of apoptosis was observed in Cucurbitacin D treated cervical cancer cells as measured by enhanced Annexin V staining and cleavage in PARP protein. Cucurbitacin D treatment of cervical cancer cells arrested the cell cycle in G1/S phase, inhibited constitutive expression of E6, Cyclin D1, CDK4, pRb, and Rb and induced the protein levels of p21 and p27. Cucurbitacin D also inhibited phosphorylation of STAT3 at Ser727 and Tyr705 residues as well as its downstream target genes c-Myc, and MMP9. Cucurbitacin D enhanced the expression of tumor suppressor microRNAs (miR-145, miRNA-143, and miRNA34a) in cervical cancer cells. Cucurbitacin D treatment (1 mg/kg body weight) effectively inhibited growth of cervical cancer cells derived orthotopic xenograft tumors in athymic nude mice. These results demonstrate the potential therapeutic efficacy of Cucurbitacin D against cervical cancer.

Summary: A novel bis(2-thienyl)disulfide derivative was synthesized through a reaction involving ethyl 3-oxobutanoate, carbon disulfide, and chloroacetone. The synthesized compound was characterized by spectral and X-ray analyses, and the results were confirmed using density functional theory calculations. Additionally, in silico studies were conducted to elucidate the inhibitory mechanism of the compound against dihydrofolate reductase.

Synthesis and Biological Evaluation of Certain Tetrasubstituted Thiophene Derivatives

Summary: A series of thiophene derivatives containing pyrazolo[3,4-d]pyridazine and pyridine moieties were synthesized and characterized. The synthesized compounds showed in vitro antimicrobial activity against various bacteria and fungi, with compounds 6 and 7 exhibiting the highest activity. Compound 6 demonstrated potent antibacterial activity against P. aeruginosa and moderate cytotoxic activity against the colon carcinoma cell line HCT-116.

Summary: There is an urgent need for the development of new antimicrobial drugs to combat the emergence of antimicrobial-resistant microbes. A series of new thiophene derivatives were synthesized and tested for their antibacterial and antifungal activities. The compounds showed significant antibacterial activity against Gram-positive strains and strong antifungal activity. The results provide a foundation for the development of lead compounds with improved antimicrobial activities.

Intravenous Nanocarrier for Improved Efficacy of Quercetin and Curcumin against Breast Cancer Cells: Development and Comparison of Single and Dual Drug-Loaded Formulations Using Hemolysis, Cytotoxicity and Cellular Uptake Studies

Summary: This study highlights the suitability of an oil-based nanocarrier for delivering quercetin and curcumin intravenously for the treatment of breast cancer. The nanoemulsion showed favorable characteristics such as small particle size, low polydispersity index, and suitable viscosity. In vitro experiments demonstrated the biocompatibility of the nanoemulsion with human blood and its cytotoxic potential against breast cancer cells, with a synergistic effect observed when both drugs were combined. The cellular uptake of the loaded nanoemulsion was significantly higher than that of the free drugs. Overall, this strategically designed nanoemulsion appears to be a promising drug delivery system for the development of quercetin and curcumin as therapeutic modalities for breast cancer treatment.

In Vitro, Molecular Docking and In Silico ADME/Tox Studies of Emodin and Chrysophanol against Human Colorectal and Cervical Carcinoma

Summary: In this study, anthraquinone derivatives chrysophanol (Chr) and emodin (EM) showed strong anti-cancer effects and had better binding energies than the FDA-approved anticancer drug fluorouracil, according to molecular docking studies. In vitro experiments confirmed the promising anticancer activity of Chr and EM in HCT-116 and HeLa cells, laying the foundation for their potential use in the treatment of human colorectal and cervical carcinomas.

The results showed that patients were highly satisfied with the accessibility, facilities, and staff of the pharmacies. They also had a positive experience with the new service compared to the old pharmaceutical services at primary healthcare centers.

The residents demonstrated a high level of awareness regarding complementary and alternative medicine, but there is a need for increased awareness in individuals using such therapies for progressive medical conditions.

Using Design Thinking Method in Academic Advising: A Case Study in a College of Pharmacy in Saudi Arabia

Summary: The current study aimed to evaluate academic advising services in a pharmacy college in Saudi Arabia and develop solutions to overcome the identified challenges. Using the design thinking method, several issues were identified with students, and prototypes were suggested to enhance their experiences in university life. These prototypes include a peer academic advising club, personal development workshop, and a series of lectures on college rules and regulations.
