(+)- and (-)-Ganodilactone (1), a pair of novel meroterpenoid dimers possessing a unique 5'H-spiro [chroman-4,2'-furan]-2,5'-dione ring system, along with their biogeneticly-related compound, ganomycin I (2), were discovered from the fruiting bodies of Ganoderma leucocontextum. The structures and absolute configurations were assigned with the aid of 1D and 2D NMR spectra, HRESIMS analysis and ECD calculations. (+/-)-, (+)-, and (-)-ganodilactone (1) showed pancreatic lipase inhibitory activities and exhibited the IC50 values as 27.3, 4.0, and 2.5 mu M, respectively.

Summary: A photoinduced intramolecular cyclization of allyl acrylates to synthesize fully substituted spiro-gamma-butyrolactone compounds is described. The reaction is believed to proceed through activation of the carbonyl group upon ultraviolet irradiation, 1,4-hydrogen atom transfer, and intramolecular cyclization of diradical species, an intramolecular hydrogen abstraction strategy, based on a series of control and isotope studies as well as DFT calculations.

Sesquiterpenes with diverse skeletons from histone deacetylase inhibitor modified cultures of the basidiomycete Cyathus stercoreus (Schwein.) De Toni HFG134

Summary: Epigenetic modifiers have been found to regulate the gene expression of fungal biosynthetic pathways. In this study, a chemical investigation on histone deacetylase inhibitor (HDI)-modified cultures of Cyathus stercoreus led to the discovery of nine previously undescribed sesquiterpenes. These sesquiterpenes were found to be exclusive or overproduced in the epigenetic modified cultures. Additionally, a previously unreported sesquiterpene skeleton was identified. Some of the compounds showed weak inhibitory activity against Escherichia coli.

Structure elucidation of linear triquinane sesquiterpenoids, hirsutuminoids A-Q, from the fungus Stereum hirsutum and their activities

Summary: Eighteen linear triquinane sesquiterpenoids, including seventeen previously undescribed ones, were isolated from the fungus Stereum hirsutum. The structures and absolute configurations of the isolates were determined through extensive spectroscopic analysis and X-ray crystallography. The study also summarized an empirical rule about the orientations of substitutions at specific positions. Among the compounds, hirsutuminoid B showed anti-inflammatory and cytotoxic activity.

Tetra-, penta-, and hexa-nor-lanostane triterpenes from the medicinal fungus Ganoderma australe

Summary: Ten new nor-lanostane triterpenes were identified from the medicinal fungus Ganoderma australe through chemical investigation. The chemical structures and absolute configurations were determined, and the biosynthesis pathways of these compounds were proposed. Two of the compounds showed moderate inhibition against nitric oxide production in macrophage cells.

Lanostane triterpenoids from mycelial cultures of the basidiomycete Ganoderma weberianum

Summary: Eight undescribed lanostane triterpenoids (1-8) were isolated from cultures of Ganoderma weberianum TBRC-BCC 60642. Lanostane constituents showed different structural types between mycelial cultures and fruiting bodies. Lanostanes 2, 3, and 6 exhibited moderate antimalarial activity against Plasmodium falciparum K1.

Cordycicadins A-D, Antifeedant Polyketides from the Entomopathogenic Fungus Cordyceps cicadae JXCH1

Nine New Pregnane Glycosides from the Cultivated Medicinal Plant Marsdenia tenacissima

Summary: In this study, nine new pregnane glycosides were isolated from the ethnobotanical plant Marsdenia tenacissima, commonly used by the Dai people in Yunnan Province, China. The structures were analyzed using spectroscopic techniques and the absolute configurations of the sugar moieties were determined. Some of the isolated compounds showed inhibitory activity against nitric oxide production in activated macrophage cells.

Triquinane Sesquiterpene Glycosides from the Basidiomycete Antrodiella zonata

Summary: In our study, we isolated twelve previously undescribed triquinane sesquiterpene glycosides, antrodizonatins A-L (1-12), and four known compounds (13-16) from the fermentation of the basidiomycete Antrodiella zonata. The structures of these compounds were determined through extensive spectroscopic analysis and theoretical calculations. Furthermore, compounds 1, 5, and 12 exhibited antibacterial activity against Staphylococcus aureus with MIC50 values of 35, 34, and 69 μM, respectively.

Three previously undescribed metabolites from Cordyceps cicadae JXCH-1, an entomopathogenic fungus

Summary: Three previously undescribed compounds were isolated from the cultures of Cordyceps cicadae JXCH1. Their structures and relative configurations were determined by NMR spectroscopic analysis and crystal structure determination. One of the compounds exhibited an unusual structural feature and showed inhibitory effects on human lung cancer cells and murine macrophages.

Structure-based redesign of Fe(ii)/2-oxoglutarate-dependent oxygenase AndA to catalyze spiro-ring formation

Summary: Structure- and mechanism-based redesign of the Fe(ii)/2-oxoglutarate-dependent oxygenase AndA was performed to expand its function from an isomerization reaction to catalyzing a spiro-ring formation reaction. The redesigned AndA variants produced two unnatural novel spiro-ring containing compounds through two and three consecutive oxidation reactions.
