Expressions of Matrix Metalloproteinases (MMP-2, MMP-7, and MMP-9) and Their Inhibitors (TIMP-1, TIMP-2) in Inflammatory Bowel Diseases

Crohn's disease (CD) and ulcerative colitis (UC) belong to a group of inflammatory bowel diseases (IBD). The aim of our study was to evaluate the expression of MMP-2, MMP-7, MMP-9, TIMP-1, and TIMP-2 in ulcerative colitis and Crohn's disease. The study group comprised 34 patients with UC and 10 patients with CD. Evaluation of MMP-2, MMP-7, MMP-9, TIMP-1, and TIMP-2 expression in tissue samples was performed using immunohistochemistry. The overexpression of MMP-9 and TIMP-1 was dominant in both the glandular epithelium and inflammatory infiltration in UC patients. In contrast, in CD subjects the positive expression of MMP-2 and TIMP-1 was in glandular tubes while mainly MMP-7 and TIMP-2 expression was in inflammatory infiltration. Metalloproteinases' expression was associated with the presence of erosions, architectural tissue changes, and inflammatory infiltration in the lamina propria of UC patients. The expression of metalloproteinase inhibitors correlated with the presence of eosinophils and neutrophils in UC and granulomas in CD patients. Our studies indicate that the overexpression of metalloproteinases and weaker expression of their inhibitors may determine the development of IBD. It appears that MMP-2, MMP-7, and MMP-9 may be a potential therapeutic target and the use of their inhibitors may significantly reduce UC progression.

Summary: The study aimed to determine the clinical usefulness of NETs biomarkers in saliva as a potential prognostic and therapeutic target in patients with oral squamous cell carcinoma (OSCC). Results showed significantly lower concentrations of NOX1, NCF2, and DNASE1 in the saliva samples of cancer patients, suggesting an impairment of immunological homeostasis within the oral cavity.

Are Matrix Metalloproteinase-9 and Tissue Inhibitor of Metalloproteinase-1 Useful as Markers in Diagnostic Management of Children with Newly Diagnosed Ulcerative Colitis?

Summary: This study aimed to investigate the relationship between MMP-9 and TIMP-1 and UC phenotypes, disease activity index, and inflammatory markers. The results showed that children with UC had significantly higher levels of these markers in their serum and feces compared to the control group. Additionally, serum MMP-9 correlated with the severity of the disease and other markers, while serum TIMP-1 correlated with disease activity, inflammatory markers, and erythrocyte sedimentation rate. Serum and fecal levels of MMP-9 and TIMP-1 can be useful in distinguishing UC patients and evaluating disease phenotypes.

Establishment of In Vitro and In Vivo Anticolorectal Cancer Efficacy of Lithocholic Acid-Based Imidazolium Salts

Summary: This research evaluated a series of novel imidazolium salts substituted with lithocholic acid and alkyl chains of various lengths against colon cancer cells. The results showed that these salts have anticancer activity, which is related to the length of the alkyl chain. The study also demonstrated the inhibitory effect of one compound on tumor growth in a mouse model.

Beneficial Proapoptotic Effect of Heterobasidion Annosum Extract in Colorectal Cancer Xenograft Mouse Model

Summary: Fungal extract from Heterobasidion annosum (HA) has been evaluated for its potential anticancer activity in colorectal cancer. In an in vivo model, mice with implanted DLD-1 human cancer cells were treated with HA extract, the drug 5-fluorouracil (5FU), or a combination of both. Results showed that the combination treatment led to the smallest tumor volumes, with increased concentrations of apoptotic proteins caspase-8 and p53, and decreased levels of survivin and Bcl-2. The Ki-67 protein was found to be expressed in all tested groups. These findings provide innovative insights into the antitumor activity of HA extract and its interaction with chemotherapeutic agents.

Simultaneous analysis of tumor-infiltrating immune cells density, tumor budding status, and presence of lymphoid follicles in CRC tissue

Summary: This study conducted a simultaneous analysis of tumor-infiltrating immune cell density, presence of lymphoid follicles, and budding status in colorectal cancer tissue for the first time. The results showed that tumor budding, lymphocyte infiltration, and lymphoid follicles could serve as reliable indicators of colorectal cancer progression.

The caspase-8 and procaspase-3 expression in gastric cancer and non-cancer mucosa in relation to clinico-morphological factors and some apoptosis-associated proteins

Summary: The aim of this study was to assess the expression of caspase-8 and procaspase-3 proteins in gastric cancer cells and non-cancerous mucosa, and its relationship with clinical and morphological characteristics of the tumor, postoperative survival, and other apoptosis-related proteins. The results showed that procaspase-3 was expressed in all gastric cancer cells, with a significant difference in expression compared to non-cancerous mucosa. Caspase-8 expression was more common in certain types of gastric cancer and was associated with longer survival and better prognosis.

E-Cadherin Expression Varies Depending on the Location within the Primary Tumor and Is Higher in Colorectal Cancer with Lymphoid Follicles

Summary: Different distributions of E-cadherin expression were observed within colorectal cancer, with the highest percentage of positive expression in the invasive front and tumor center. The expression pattern varied depending on cancer progression and other clinical and histopathological features. Low E-cadherin expression appears to be a reliable indicator of higher cancer staging and progression.

Summary: Magnification endoscopy and methylene blue chromoendoscopy can effectively detect intestinal metaplasia (IM). Methylene blue staining is more accurate in indicating IM compared to pit pattern and vessel evaluation. Methylene blue chromoendoscopy can serve as a screening method for detecting advanced OLGIM stages.

Diagnostic significance and utility of circulating redox biomarkers in patients with gastric cancer - preliminary study

Summary: Gastric cancer is associated with redox imbalance and increased oxidative damage to proteins, lipids, and DNA. Histopathological parameters of the tumor are associated with the level of antioxidants and oxidative damage products. Determination of redox parameters may be useful in the assessment of the tumor progression.

May the Nitrosative and Carbonyl Stress Promote Inflammation in Patients with Colorectal Cancer?

Summary: The study evaluated the differences in nitrosative stress, protein and DNA oxidation/glycoxidation, as well as pro- and anti-inflammatory cytokines between CRC patients and healthy controls. The results showed that oxidation and glycoxidation products in CRC patients were positively correlated with pro-inflammatory and anti-inflammatory cytokines, indicating that redox damages may promote inflammation in CRC.
